上皮生长因子对小鼠的致畸胎作用研究

Teratology studies on epidermal growth factor in mice

摘要: 目的 了解上皮生长因子(epidermal growth factor,EGF)的致畸作用和胚胎毒性作用。方法 EGF 设2,12,72μg·Kg-13 个剂量组,分别相当于人临床拟用剂量的3,18,108 倍。于小鼠妊娠第6 ~15 d 连续灌胃给药,每天1 次。结果 EGF 72μg·kg-1对小鼠活胎率(49 %)和吸收胎率(50 %)有影响,与溶剂对照组比较有显著性差异(P<0.05)。2和12μg·kg-1组及溶剂对照组的活胎率、吸收胎率、死胎率和骨骼畸形率均在正常范围,与各给药组比较无显著的统计学意义(P>0.05)。阳性对照组用环磷酰胺20 mg·kg-1,外观畸形率达90 %,表明本次试验结果可靠。结论 EGF 相当于人临床拟用剂量的108 倍有明显的胚胎毒性,未见致畸胎作用。

Abstract: AIM: To observe the teratology of epidermal growth factor (EGF)orally administered in mice.METHODS: Doses of EGF 2,12,and 72μg·kg-1·d-1were given by iPon d 6-15 of in mice with gestation.RESULTS: Fetuses were removed by cesarean section 1dPrior to expectedParturition,the rates of viable fetuses,absorption,dead fetuses,external and skeletal abnormality were recorded.Compared with vehicle control group,the rates of viable fetuses and absorption increased markedly in 72μg·kg-1treatment group.CONCLUSION: The epidermal growth factor shows embryotoxicity,but no evidence of teratogenicity.
