Effects of rAAV/rSTAP on mRNA level of interstitial collagenase in rat stellate cells

摘要: 目的: 研究重组副腺病毒介导的大鼠星状细胞激活相关蛋白(stellate cell activation-associated Protein, STAP) 基因 转染对 大鼠肝 星状细 胞表达 间质肢原酶mRNA的影响。方法: 构建并制备rAAV/iSTAP病毒载体。链霉蛋白酶、胶原酶原位灌注法分离纯化SD大鼠肝星状细胞,转染HSC,用半定量RT-PCR法研究对肝星状细胞表达间质胶原酶mRNA的影响。结果: rAAV/rSTAP转染可提高大鼠肝星状细胞表达间质胶原酶Mma的水平。结论: 间质胶原酶与肝纤维化形成密切相关,rAAV/rSTAP转染可提高大鼠肝星状细胞表达间质胶原酶mRNA的水平,提示rAAV/rSTAP上调大鼠肝星状细胞间质胶原酶mRNA的表达水平可能是其抗纤维化的机理之一。

Abstract: AIM: To observe tlie effects of rAAV/ rSTAP on mRNA level of interstitial collagenase in rat stellate cell. METHODS: Recombinant adeno-associated virus including STAP gene was constructed and obtained. The stellate cells were isolated from SD rat5 s liver and cultured in DMEM medium. The effects of STAP on the level of interstitial collagenase were measured by RT-PCR. RESULTS: The level of interstitial collagenase in rAAV/rSTAP treated cells increased significantly com-pared with that in control group. CONCLUSION: rAAV/rSTAP can up regulate the expressions of interstitial collagenase mRNA and may be associated with anti-hepat-ic fibrosis.
