Effects of ecdysterone on injury of endothelial cells following experimental subarachnoid hemorrhage

摘要: 目的 观察血性脑脊液(BCSF) 对培养的微血管内皮细胞(brain microvessel endothelial cells, BmvECs)损害作用和蜕皮甾酮(ecdysterone, EDS) 的干预效应。 方法 将培养的BmvECs 分为对照组、BCSF组和EDS 组, 通过病理形态学改变、细胞计数、MTT比色法和流式细胞仪细胞周期分析评估其损害和增殖状况。 结果 与对照组比较, BCSF 组内皮细胞出现明显的病理形态学改变, 贴壁细胞数量减少, MTT吸光度下降,G0 ~ G1期细胞比例减少。EDS 对BCSF所致上述内皮细胞改变有一定的改善作用。 结论 EDS 对BCSF 所致的内皮形态和增殖损害具有保护作用, 可能在慢性脑血管痉挛治疗中具有一定作用。

Abstract: AIM: To investigate the effects of ecdysterone (EDS) on injury and proliferation of endothelial cells caused by bloody cerebralspinal fluid (BCSF). METHODS: Rabbit brain microvessel endothelial cells (BmvECs) were divided into control group, BCSF group and EDS group, which were treated with DMEM, BCSF and both of BCSF and EDS (200 mg·L-1), respectively. Morphological changes, cell counting, MTT assay, and flow cytometric analysis were used to evaluate the cytotoxic effects on BmvECs. RESULTS: BCSF could cause severe morphologic changes in BmvECs, decrease the OD of MTT assay and cell proportion of G0-G1 of BmvECs(P <0.01).EDS could alleviate these changes in BmvECs caused by BCSF. CONCLUSION: EDS has protective effects on the injury of BmvECs caused by BCSF, which suggests EDS may play a role in the therapy of CVS.
