积雪草苷合大黄素对肿瘤坏死因子α诱导的肾系膜细胞C3表达的影响

Effects of Centalla asiatica combining emodin on complement C3 in glomerular mesangial cells stimulated by Tumor Necrosis Factor α

摘要: 目的:探讨积雪草苷合大黄素干预肿瘤坏死因子α(TNFα)诱导的BALB/C 小鼠肾小球系膜细胞补体核心成分-C3 mRNA 及蛋白的表达水平。方法:采用BALB/C 第3 ~ 5 代肾小球系膜细胞, 模型组TNFα5 ng·ml-1诱导24 h, 治疗组在TNFα诱导的同时, 分别以积雪草苷1 、2 、4 μg·ml-1合大黄素0.1 、0.2 、0.4 μg·ml-1进行干预, 于24 h 后分别提取细胞上清及RNA, 应用逆转录-聚合酶链反应(RT-PCR)和酶链免疫方法(ELISA)分别检测肾小球系膜细胞C3mRNA 和蛋白的表达。结果:正常BALB/C 小鼠的肾小球系膜细胞具有一定的C3 mRNA 和蛋白表达。经TNFα诱导后C3 mRNA 和蛋白表达均明显上调, 用上述3 种不同浓度的积雪草苷合大黄素干预后, 肾小球系膜细胞C3 mRNA 及蛋白水平表达均出现不同程度的下调, 呈一定的剂量依赖关系。结论:积雪草苷合大黄素能够抑制炎性细胞因子TNFα上调所致的肾局部C3 过度产生, 具有保护肾功能、延缓病程进展的作用。

Abstract: AIM: To investigate the effects of Centalla asiatica combining emodin on complement C3 mRNA and protein expression in BALB/C mice glomerular mesangial cells (MsC)stimulated by Tumor Necrosis Factor α(TNFα).METHODS: Cultured mice MsC from thirdfifth generation were exposed to TNFα5 ng·ml -1 and Centella asiatica (1, 2 and 4 μg·ml -1)combining emodin (0.1, 0.2 and 0.4 μg·ml -1) for 24 h.The transcriptase-polymerase chain reaction (RT-PCR)and enzyme-linked immunosorbent assay (ELISA)were performed to analysis complement C3 mRNA and protein expression. RESULTS: TNFαup-regulated C3 expression by MsC. This effect was inhibited by different concentrations of Centalla asiatica combining emodin in a dose-dependent manner.CONCLUSION: Centalla asiatica combining emodin inhibits C3 expression stimulated by TNFαin source of kidney local.It can protect function of kidney and delay development of nephropathic process.
