白藜芦醇诱导K562 细胞凋亡过程中Bcl-2蛋白水平的改变

Changes of Bcl-2 expression in apoptosis induced by resveratrol in K562 cells

摘要: 目的:观察白藜芦醇对K562 细胞的诱导凋亡效应, 探讨其可能的作用机制。方法:应用噻唑蓝(MTT) 比色法、光镜、电镜观察K562 细胞凋亡;流式细胞术(FCM) 测定K562 细胞Bcl-2 、P53 蛋白水平的变化;比色法检测Caspase-3 活性变化。结果:白藜芦醇可抑制K562 细胞增殖, 光镜和电镜下观察呈典型凋亡形态改变, Bcl-2 蛋白表达明显下调, Caspase-3 活性明显增强。结论:白藜芦醇可通过Bcl-2 依赖性Caspase-3 激活而诱导K562 细胞凋亡。

Abstract: AIM: To observe the apoptosis of K562 cells induced by resveratrol and to explore its possible mechanisms.METHODS: MTT assay, light microscopy and electron microscopy were used to examine apoptosis in K562 cells.Expression levels of Bcl-2 and P53 antigens were mesured by FCM.Colorimetric assay was employed to detect the activation of Caspase-3.RESULTS: Resveratrol could inhibit growth of K562 cells.Typical morphological changes of apoptosis were observed through light microscopy and electron microcopy.Expression of Bcl-2 significantly decreased after application of resveratrol.Caspase-3 could also be activated by resveratrol. CONCLUSION: Resveratrol induces apoptosis in K562 cells by activating Caspase-3 via a Bcl-2-dependent pathway.
