A 型肉毒毒素降低偏头痛大鼠颈静脉血、脑干和三叉神经节的P物质含量

Botulinum toxin type A reducing SPimmunoreactivity in jugular plasma,brain stem and ganglion nervi trigemini in migraine model rats triggered by nitroglycerin

摘要: 目的 通过观察A 型肉毒毒素(botulinum toxin type A, BTX-A) 对硝酸甘油引发的偏头痛大鼠颈静脉血浆、脑干和三叉神经节中P物质(substance P,SP) 含量变化, 探讨BTX-A 止痛机理。方法 将大鼠分为对照组(n =5) 和偏头痛模型组(n =15) 。偏头痛模型组造模后分别皮下注0.9%NaCl (n =5) 、BTX-A 5 U·kg-1 (n =5) 、BTX-A 10 U·kg-1 (n =5)6 d 后, 应用放射免疫检测法测定颈静脉血浆、脑干和三叉神经节中P物质的含量。结果 动物皮下注射硝酸甘油后出现偏头痛症状, 例如双耳发红、前肢频繁搔头、爬笼次数增多, 其评分与对照组相比有明显差异(P<0.01) 。生理盐水组颈静脉血浆、脑干和三叉神经节中SP含量与对照组比较显著升高(P<0.01);BTX-A 5 U·kg-1 组、BTX-A 10 U·kg-1 组分别与生理盐水组比较, SP含量显著降低(P<0.05) 。结论 硝酸甘油导致偏头痛症状产生及SP在颈静脉血浆和延髓、脑桥和三叉神经节中的含量增加。局部皮下注射BTX-A 可减轻偏头痛大鼠的症状, 并能降低颈静脉血浆、脑干和三叉神经节中SP含量, 结果提示BTX-A 通过抑制“三叉神经血管系统”(trigeminalvascular system, TVS) 中SP释放而减轻偏头痛症状。

Abstract: AIM: To observe the content change of substance P(SP) in jugular plasma, brain stem and ganglion nervi trigemini followed botulinum toxin type A(BTX-A) injected subcutaneously into migraine model rats triggered by nitroglycerin and its mechanism.METHODS: Rats were divided randomly into control grouP(n =5) and migraine model grouP(n =15) triggered by nitroglycerin injected local subcutaneously,which was subdivide into grouPtreated with NS (n =5) ,grouPtreated with BTX-A 5 U·kg-1 (n =5) and grouPtreated with BTX-A 10 U·kg-1 (n =5).The BTX-A and NS were injected into frontal and temporal for 6 days.The immunoractive SP(iSP) in jugular plasma, brain stem and ganglion nervi trigemini was detected after migraine treatment by RIA.RESULTS: Subcutaneous injection of nitroglycerin into animal caused symptoms of migraine, such as ear rosacea, forelimb clawing head frequently and climbing cage more, and behavioral scores increasing (P<0.01).The content of iSPin jugular plasma, brain stem and ganglion nervi trigemini was increased signify cantly in NS grouPthan that in control grouP(P<0.01).The contents of iSPin jugular plasma of BTX-A 5U·kg-1 and BTX-A 10 U·kg-1 groups decreased significantly(P<0.05) than that in NS group.CONCLUSION: Nitroglycerin subcutaneous injection triggers symptoms of migraine and increases content of SPin jugular plasma, brain stem and ganglion nervi trigemini.BTX-A local subcutaneous injection reduces symptoms of migraine model rats and content of SPin jugular plasma,brain stem and ganglion nervi trigemini.The results suggest that BTX-A can inhibit SPreleasing from “trigeminal vascular system”and prevent migraine from meningeal sterile inflammatory response.
