华蟾素对耐阿霉素人乳腺癌细胞内阿霉素蓄积的影响

Effects of cinobufacine on intracellular adriamycin accumulation in human breast cancer cells

摘要: 目的: 测定华蟾素(cinobufacine,Cino)对耐阿霉素人乳腺癌细胞MCF-7/ADM 内阿霉素(ADM)积累的影响,以探索Cino 逆转MCF-7/ADM 多药耐药(MDR)的可能机制。方法: MTT 法检测Cino MDR 的逆转作用,HPLC 法检测Cino 作用MCF-7/ADM 后细胞内ADM 的浓度的变化。结果: Cino 15 mg/L 能增加MCF-7/ADM 细胞对ADM 的敏感性,使ADM 的半数抑制浓度(IC50)由38.14 mg/L 降至12.93 mg/L,显著增加MCF-7/ADM 株细胞内ADM 的浓度。结论: Cino 能明显增加MCF-7/ADM 株细胞内ADM 的含量,部分逆转MCF-7/ADM 的MDR。

Abstract: AIM: To study the effects of cinobufacine on intracellular adriamycin accumulation in human breast cancer MCF-7/ADM cells, and further explore the mechanism of cinobufacine reversing the multidrug resistance of human breast cancer MCF-7/ADM cells. METHODS: The cytotoxic effect was determined by MTT assay. Intracellular ADM concentration was examined by HPLC. RESULTS: 15 mg/L cinobufacine decreased the IC50 of ADM to MCF-7/ADM cells from 38.14 mg/L to 12.93 mg L, but increased intracellular adriamycin accumulation in MCF-7/ADM cells. CONCLUSION: Cinobufacine can increase intracellular ADM accumulation and partly reverse MDR of MCF-7/ADM cell line.
