冠欣舒胶囊对大鼠心肌细胞缺氧/复氧损伤保护作用的拆方实验研究

Experiment study of Guanxinshu capsule attenuates anoxia/reoxygenation injury in neonatal rat cardiomyocytes by partial formulation analysis

摘要: 目的: 研究冠欣舒胶囊对抗心肌细胞缺氧/复氧(A/R)损伤作用,通过正交设计进行拆方实验,以求其最佳配比方案。方法: 按原方中丹参、三七、降香分离所得的有效部位及基本配比,设计正交表L16(44);取培养4 d的大鼠乳鼠心肌细胞,随机分为18组(n=8):(1) Cont组、(2)A/R组、(3)~(18)受试组进行实验;检测细胞存活率、超氧化物歧化酶(SOD)、Catalase、谷胱甘肽过氧化物酶(GPx)活性、丙二醛(MDA)含量及孵育液乳酸脱氧酶(LDH)活性;用综合平衡法进行分析,得出冠欣舒最佳配比方案,并进行验证实验。结果: 不同配比组合的冠欣舒处理组可使细胞存活率与SOD、Catalase、GPx活性均有不同程度的升高,LDH活性与MDA含量均有不同程度的降低(P<0.01或P<0.05);三七总皂苷与丹参总酚酸分别在保护细胞与提高细胞抗脂质过氧化作用方面发挥重要作用;冠欣舒的最佳配比组合为:A3B3C2D4结论: 冠欣舒不同的配比具有对抗心肌细胞A/R损伤的作用,其机制可能与抗脂质过氧化作用有关;冠欣舒的最佳配比组合为丹参总酚酸0.027 mg、三七总皂苷0.048 mg、降香油0.002625 mg、降香总黄酮0.01326 mg。

Abstract: AIM: To investigate the protective effect of guanxinshu capsule on cardiomyocytes after anoxia-reoxygenation(A/R) injury, partial formulation experiment was executed by orthogonal design in order to obtain the best ratio of Guanxinshu.METHODS: Based on the effective site and the basic ratio isolated from original chemical formulation of salvia miltiorrhiza, panax pseudo-ginseng, lignum acronylchiae, orthogonal table was designed.The primarily neonatal rat ventricular cardiomyocytes were randomly divided into 18 groups: (1)control group; (2)A/R group; (3)-(18) experimental groups treated with different ratio; Cell viability, SOD, Catalase, GPx activities, MDA contents and LDH activitied were detected.Best ratio of program was obtained by analysis of integrated equilibrium and then verificated.RESULTS: These groups treated with different ratio of Guanxinshu elevated cell viability and SOD, Catalase, GPx activity, whereas LDH activities and MDA contents reduced(P<0.01 or P<0.05); Panax pseudo-ginseng saponins and salvia miltiorrhiza phenolic acid have played an important role of protection of cardiomyocyte and increased ability of attenuation lipid peroxidation; The best ratio of Guanxinshu is: A3B3C2D4.CONCLUSION: The different ratio of Guanxinshu have protected cardiomyocyte against A/R injury, the mechanism may be relative with attenuation lipid peroxidation.The best ratio of Guanxinshu is: salvia miltiorrhiza total phenolic acid 0.027 mg, Panax pseudo-ginseng total saponins 0.048 mg, lignum acronylchiae oil 0.002625 mg, lignum acronylchiae total flavonoids 0.01326 mg.
