μ阿片受体基因内含子2多态性与美沙酮维持治疗剂量的关联分析

摘要: 目的:探讨μ阿片受体基因内含子2的两个单核苷酸多态性位点(Single nucleotide polymorphism, SNP) rs9479757和rs2075572与海洛因成瘾者美沙酮维持治疗剂量的相关性。方法:应用TaqMan荧光技术检测68例美沙酮维持治疗病例的rs9479757和rs2075572的基因型并进行与美沙酮维持剂量高低的关联分析。结果:rs9479757和rs2075572在美沙酮维持治疗高、低剂量组间的分布差异无统计学意义;rs9479757和rs2075572各基因型与美沙酮维持治疗剂量差异无统计学意义。结论:μ阿片受体基因内含子2的两个SNP (rs9479757和rs2075572)多态性与海洛因成瘾者美沙酮维持剂量的高低无明显相关性。

Abstract: AIM: To explore the association between the single nucleotide polymorphisms (SNP) in intron 2 of the μ opioid receptor gene (OPRM1) and the effective dose in methadone maintenance treatment of heroin addiction.METHODS: The genotypes of the two SNPs (rs9479757, rs2075572) in 68 heroin dependent patients in methadone maintenance treatment were detected by TaqMan SNP genotyping technique, and the association between methadone dose and the two SNPs was analyzed.RESULTS: The frequencies of genotype and allele at rs9479757, rs2075572 were not significantly different between the high and low methadone dose groups (P>0.05). The average methadone daily dosages of various genotypes at the two SNPs were not significantly different (P>0.05).CONCLUSION: The polymorphisms (rs9479757, rs2075572) in intron 2 of the μ opioid receptor gene (OPRM1) may not play the major role in the effective dose in methadone maintenance treatment of heroin addiction.
