茯苓对胃癌细胞株SGC-7901的侧群细胞增殖的影响

Effect of poria on the proliferation of the side population of gastric cancer cell line SGC-7901

摘要: 目的 探讨含茯苓药物血清对胃癌细胞SGC-7901侧群细胞(side population,SP)的增殖能力的影响。方法 首先制作含茯苓药物血清。之后应用流式细胞仪技术分选体外培养的胃癌SGC-7901细胞株中的SP细胞。应用CCK-8法观察含茯苓药物血清干预SP细胞后细胞增殖能力的变化。之后利用流式细胞仪技术检测含茯苓药物血清干预SP细胞后细胞周期的变化。结果 胃癌SGC-7901细胞株中SP细胞比例为 4.4%±0.6%。含茯苓药物血清干预后,胃癌SGC-7901细胞株SP细胞的增殖能力受到明显抑制(P<0.01)。此外,含茯苓药物血清干预后,SP细胞的细胞周期被阻滞于G0、G1期,而S、G2/M期细胞减少,并且随着干预时间的延长,此种趋势更加明显,同时也就验证了细胞增殖实验的实验结果。结论 研究结果表明,胃癌SGC-7901细胞株中存在且能成功分选出SP细胞。含茯苓药物血清通过对细胞周期的改变,来抑制胃癌SGC-7901细胞株SP细胞的增殖能力。

Abstract: AIM: To investigate the effect of poria drug serum on the proliferation of the side population (SP) of gastric cancer cell line SGC-7901. METHODS: The making of poria rabbit serum drug; Sorting the gastric cancer cell line SGC-7901 SP cells by flow cytometry technique; Observe the effect of poria drug serum on the proliferation of the gastric cancer cell line SGC-7901 SP cells by CCK-8; Observe the effect of potia drug serum on the cell cycle of the gastric cancer cell line SGC-7901 SP cells by flow cytometry technique. RESULTS: The percentage of side group in gastric cancer cell line SGC-7901 was 4.2%±0.6%; Gastric cancer cell line SGC-7901 SP cells proliferation ability was significantly inhibited by poria drug serum; The cell cycle of SP cells was blocked in G0, G1 phase while the S and G2/M phase cells were on the decrease after the intervention of poria drug serum. At the same time, longer duration of intervention, the trend was more obvious, so the experimental results of cell proliferation was verified. CONCLUSION: SP cells can be successfully sorted out of gastric cancer SGC-7901 cell lines. With poria drug serum on the change of cell cycle, the proliferation of the gastric cancer cell line SGC-7901 SP cells can be suppressed.
