地塞米松对急性胰腺炎小鼠胰腺组织中NF-κB及survivin表达的影响

Effects of dexamethasone on the expression of NF-κB and survivin in acute pancreatitis

摘要:

目的: 探讨地塞米松对急性胰腺炎小鼠胰腺组织中NF-κB及survivin表达的影响。方法: 利用蛙皮素建立急性胰腺炎(acute pancreatitis,AP)动物模型,实验分组为生理盐水组(对照组)、AP组、地塞米松(DEX)组,利用ELISA法检测各组动物血清中TNFα的含量;利用实时荧光定量PCR(Q-PCR)与western blot检测各组胰腺组织中NF-κB、survivin、TNF-α基因与蛋白的表达情况。 结果: ELISA检测结果显示,与AP组相比较,DEX组动物血清中TNFα的含量显著降低;Q-PCR与western blot结果显示,与AP组相比较,DEX组胰腺组织中NF-κB,survivin,TNF-α mRNA与蛋白的表达均显著降低。结论: DEX可能通过抑制NFκB的活化,进而抑制survivin的表达,抑制其抗凋亡作用,促进胰腺腺泡细胞的凋亡,减少胰腺腺泡细胞的坏死,减少炎症因子,如TNF-α等的合成与释放。

Abstract:

AIM: To explore the effect of dexamethasone on the expression of NF-κB and survivin in pancreas in acute pancreatitis (AP). METHODS: AP animal model were established using caerulein and were divided into the saline group (control group), the AP group and dexamethasone group. The ELISA method was used to detect TNF-α in serum in each group. Real-time Q-PCR and Western blot were used to detect NF-κB, survivin, TNF-α mRNA and protein expression in pancreas in each group. RESULTS: The results of ELISA test showed that dexamethasone could significantly reduce the content of TNF-α in serum compared with the AP group. The results of qPCR and Western blot showed that dexamethasone could significantly reduce the expression of survivin, NF-κB, TNF-α mRNA and protein in pancreas compared with the AP group.CONCLUSION: Through inhibiting the NF-κB activation, dexamethasone could inhibit the expression and anti-apoptotic effect of survivin, promote apoptosis, reduce necrosis of pancreatic acinar cells, and inhibit the synthesis and release of TNF-α.
