黄芩苷对离体大鼠心肌缺血的保护作用

Protective effect of baicalin on myocardial ischemia in isolated rat heart

摘要: 目的: 研究黄芩苷对离体大鼠心肌缺血的保护作用及其机制。方法: 将36 只SD 大鼠分为3 组。对照组(n=12) 灌流110 min, 模型组(n=12) 灌流25 min 缺血25 min 再灌注1 h, 给药组(n=12) 灌流20 min 后换含5 mg/L黄芩苷灌流液灌流5 min, 后同模型组。监测血流动力学及冠状动脉流出量。测定冠脉流出液中CK 及LDH 水平。心肌内p38mapk 、NF-κB 表达量及TNF-ê 、IL-6 水平分别以免疫组化及ELISA 法检测。结果: 给药组较模型组心功能明显改善;冠脉流出量增多;冠脉流出液中心肌酸磷酸激酶(CK) 、乳酸脱氢酶(LDH) 均有下降(P <0.05);心肌组织TNF-ê 及IL-6 含量降低(P < 0.05);p38mapk 、NF-κB 表达减弱。结论: 黄芩苷对离体大鼠心肌缺血具有保护作用, 可能与降低心肌p38mapk 、NF-κB 等与缺血相关的酶从而下调心肌TNF-ê 及IL-6 表达有关。

Abstract: AIM: To study the protectective effect of bacalin on myocardial ischemia in isolated rat heart and its mechanism.METHODS: Thirty six Sprague-Dawley rats were randomly divided into 3 groups.The heart of rats in control group (n=12) were perfused for 1 h and 50 min incessantly.The heart of rats in ischemia group (n=12) received 25min of perfusion first and then subjected to 25 min of global ischemia followed by 1 h reperfusion.The heart of rats in baicalin group (n=12) received 5 min perfusion of 5 mg/L) baicalin before ischemia while other procession was the same as ischemia group.The hemodynamic measurement and the coronary flow were recorded.The levels of CK and LDH in the coronary effluent were detected.The expression of p38mapk, NF-κB and the levels of TNF-ê and IL-6 in myocardium were respectively detected by immumohisto- chemical technique and ELISA.RESULT: The function of hearts treated with baicalin was improved significantly, and the level of LDH and CK of coronary effluent were decreased significantly compared with ischemia group. The level of TNF-ê and IL-6 and the expression of p38mapk and NF-κB in myocardium also decreased observably compared with ischemia group.CONCLUSION: Baicalin has a protective effect on myocardial ischemia of isolated rat heart and the effect may be due to the decrease of TNF-ê and IL-6 induced by the reduction of the expression of p38mapk and NF-κB.
