地塞米松抑制肥大细胞脱颗粒及其机制初探

Inhibition of dexamethasone on degranulation of mast cells and its mechanism

摘要: 目的 研究糖皮质激素对Ⅰ 型超敏反应中的肥大细胞脱颗粒反应是否有直接作用。方法 利用流式细胞仪技术观察地塞米松对RBL-2H3 细胞(大鼠嗜碱粒细胞)脱颗粒程度的影响,利用western blot分析ERK1/ERK2 信号的改变。结果 地塞米松预先作用于RBL-2H3 细胞,使Annexin V 标记的细胞阳性率较致敏原活化组明显下降,MAPK 活性明显降低。结论 地塞米松抑制肥大细胞脱颗粒反应,其机制与MAPK 活性有关。

Abstract: AIM: To study the direct effect of dexamethasone on the degranulation of mast cells.METHODES:The flow cytometry was used to observe the effect of dexamethasone on the degranulation of RBL-2H3 cellline(rat basophileleukemia cellline),and the changes of ERK1/ERK2 signal transduction characteristics was analyzed by western blot.RESULTS:Pre-treating RBL-2H3 cellline with dexamethasone resulted in the decrease of Annexin VPositive staining of cells and the activity of MAPK compared that of the activated group.CONCLUSION: The inhibition of dexamethasone on mast cells is associated with the activity of MAPK.
