头孢拉定对奈替米星药代动力学的影响

摘要: 目的 观察头孢拉定对奈替米星药代动力学的影响。方法 14例感染患者随机分成单用奈替米 星组(NTM)和奈替米星+头孢拉定组(NTM + CPR)。采用高效液相色谱-间接光度检测 (HPLC-IPD)法, 測定患者单剂量靜脉滴注5 mg NTM后的血清药物浓度, 并计算主要药动学参数; 同时測定尿液药物浓度及药物回收率。结果 NTM组和NTM + CPR组的T1/2p分别为2. 40± l. Olh 和 4. 33± 1. 43h(P< 0. W), AUC0-24h 63. 42± 30. 00mg/ L · h 和 78. 54± 32. 88mg/ L · h(P <0. 01), 24h尿中NTM回收率也有显著性差异。结论 NTM + CPR联用时NTM生物利用度 增高, 尿中回收率下降, 连续长期联用将导致体内蓄积。

Abstract: Aim & Methods To study the effect of cephradine on pharmacokinetic action of netilmicin sulfate, the concentrations of netilmicin sulfate in serum and urine were determined by a high performance liquid chromatography-indirect photometric determination (HPLC-IPD) method at the wavelength of 262nm after intravenous drip of 5mg/kg of netilmicin sulfate in 14 patients who had been divided into two groups, with the patients in one group simultaneously taking orally cephradine. The data obtained were fitted with a PKBP-N1 program on computer. Results The disposition was conformed to a two-compartment model with T1/2β = 2. 40±1. 01h and 4. 33±1. 43h(P<0. 01), and AUC0~24h= 63. 42±30. 00mg/h·L and 78. 54±32. 88 mg/h · L(P<0. 01) in two groups. There was a remarkable discrepancy for the recla-mation of netilmicin in urine between two groups. Conclusion The bioavailability of netilmicin was raised and reclamation in urine descended with cephradine.
