绞股蓝总皂甙对氧化修饰低密度脂蛋白损伤小牛主动脉内皮细胞的防治作用

Protective effects of gypenoside on cultured bovine aortic endothelial cells against injury induced by oxidatively modified low density lipoprotein

摘要: 目的 探讨绞股蓝总皂甙(GYP)对氧化修饰低密度脂蛋白(oxidatively modified low density lipoprotein, oxLDL)损伤小牛主动脉内皮细胞的防治作用及可能机制。 方法 在体外培养的小牛主动脉内皮细胞(bovine aortic endothelial cell, BAEC)中加入oxLDL(终浓度为0.1 g pr·L-1)及GYP 低、中、高剂量组(5.0, 10.0, 20.0 mg·L-1), 培养24 h 后, 采用细胞毒试验、丙二醛(MDA)测定及细胞计数等方法, 分别对其贴壁细胞及培养液进行检测。 结果 ①细胞毒试验发现, oxLDL 对BAEC 的生长增殖有明显的抑制作用(20.63 %), GYP 组(10.0, 20.0 mg ·L-1)为9.14 %和6.56 %(P <0.01);②加oxLDL 组和GYP组BAEC 粘附的HL60 细胞数明显多于对照组(P <0.01);③MDA 检测发现oxLDL 促进BAEC 脂质过氧化物生成, 并高于GYP 组和对照组(P <0.01)。 结论 绞股蓝总皂甙具有对氧化修饰低密度脂蛋白损伤小牛主动脉内皮细胞的防治作用。

Abstract: AIM: To study the protective effects of gypenoside (GYP)on cultured bovine aortic endothelial cells against injury induced by oxidatively modified low density lipoprotein (oxLDL). METHODS: Bovine aortic endothelial cells (BAEC)were cultured and treated with oxLDL (0.1 g pr·L -1)and the different concentrations of GYP (5.0, 10.0, and 20.0 mg·L -1)for 24 h.The cultured cells and the contents in the medium were determined by MTT colorimetric assay, TBA method, cell calculating and so on. RESULTS: The inhibitory effects were shown in the proliferation of BAEC by oxLDL (20.63 %) and GYP (10.0 and 20.0 mg · L -1) (9.14 % and 6.56 %)(P <0.01).The number of HL60 monocyte cells adhered to BAEC in oxLDL group and GYP group was markedly higher than that in control group (P <0.01).MDA content in culture media in oxLDL group was significantly higher than that in control group and GYP group (P <0.01). CONCLUSION: GYP can protect cultured bovine aortic endothelial cells from injury induced by oxidatively modified low density lipoprotein.
