Possible mechanism of mitochondria on anti-apoptotic effect of ginsenoside Rg1 on MPP+-induced cellular apoptosis

摘要: 目的 探讨人参皂甙Rg1 对MPP+诱导SHSY5Y 细胞凋亡的保护作用及其可能的机制。 方法 用吖啶橙溴化乙锭染色观察SHSY5Y 细胞凋亡率, 流式细胞仪检测线粒体跨膜电位及细胞内活性氧(ROS) 水平, Western Blot 法检测bcl-2、bax 蛋白表达水平。 结果 经10 μmol·L-1 Rg1 预处理后,MPP+诱导的SHSY5Y 细胞凋亡受到明显的抑制, 虽然线粒体跨膜电位无明显改变, 但细胞内ROS 下降, 而bcl-2 蛋白表达水平增加, bax 蛋白表达水平减少。 结论 Rg1 可抑制MPP+诱导的SHSY5Y 细胞凋亡,其作用机制可能是通过清除ROS、调节bcl-2、bax 蛋白表达水平起作用。

Abstract: AIM: To explore the possible anti-apoptotic mechanism of ginsenoside Rg1 on MPP +-induced cellular apoptosis. METHODS: The apoptosis of SHSY5Y induced by MPP+was observed by AO-EB staining.Flow cytometry was used to quantitate mitochondrial transmembrane potential (△ψm) and reactive oxygen species (ROS).Western Blot was used to detect the expression of bcl-2 and bax proteins in SHSY5Y cells. RESULTS: MPP+-induced apoptosis in SHSY5Y cells was obviously inhibited by pretreatment with 10 μmol·L-1Rg1.Although there was no difference of mitochondrial transmembrane potential between Rg1 pretreated groups and MPP+groups, the level of ROS in Rg1 pretreated groups decreased, the expression of bcl-2 protein increased and expression of bax protein decreased. CONCLUSION: Rg1 protects against MPP+-induced apoptosis in SHSY5Y cells and the effect may be attributed to its remove of ROS and its regulation of expression of bcl-2 and bax proteins.
