芪苓汤、八味地黄丸、柴苓汤抗糖尿病发病和降血糖的探讨

Effects of 3 traditional Chinese formulas of herbal medicine inPrevention and treatment of diabetic rats induced by streptozotocin

摘要: 目的 探讨中草药抗糖尿病发病和降血糖的机制。方法 将大白鼠分成5 组,芪苓汤组、八味地黄丸组、柴苓汤组,分别接受相应药物治疗;模型组接受自来水治疗。各组从实验的d 8 起腹腔内注射链脲霉素30 mg·kg-1,每天1 次,连续5 d 。另设正常对照组,不作任何处理。检测大白鼠的体重、血糖、果糖胺、胰岛素及T 淋巴细胞(W3/25 和OX8)的百分率,取胰腺组织作组织学检查。结果 用药各组的糖尿病发病率比模型组低(P<0 .05)。d 26 八味地黄丸组和柴苓汤组的血糖分别比d 15 下降25%和18%(P<0 .05)。芪苓汤组和八味地黄丸组的W3/25 细胞比率明显高于对照组。用药各组的平均胰岛面积和平均β 细胞面积较模型组大。结论 八味地黄丸、柴苓汤方剂具有降血糖作用;3 方剂不同程度降低糖尿病发病率可能与提高细胞免疫功能有关。

Abstract: AIM: To investigate the mechanisms of Chinese herbal medicine formulas(Qi-ling-tang,Ba-weidi-huang-wan and Chai-ling-tang)in thePrevention and treatment of diabetes.METHODS: Wistar rats were di-· 684· Chin J ClinPharmacol Ther 2003 Dec;8(6)vided into five groups and treated with Qi-ling-tang(the grouPQLT),Ba-wei-di-huang-wan(the grouPBWDHW),Chai-ling-tang(the grouPCLT),water(the grouPmodel)and control,respectively.The rats in groups QLT through model were injected with 30mg·kg-1/daily streptozotocin(STZ)for five days beginning on day 8.The glucose concentration,fructosamine,insulin,total-protein,albumin and thePercentage ofW3/25 and OX8 T-cells in serum as well as the body weight of the rats were measured.Tissue sections ofParaffin-embeddedPancreas of the rats were observed after staining with HE,AZAN and immunohistochemistry.RESULTS: After STZ injection,the diabetic incidence of the rats in groups QLT,BWDHW and CLT were significantlylower than that in the grouPmodel,especially in the grouPBWDHW was only 21% by study day 10.A significant decrease in blood glucose concentration was found in groups BWDHW(25%)and CLT(18%)from study day 15 to 26.ThePercentage of W3/25 cells in groups QLT and BWDHW was higher than that in the control group.The average area of islets and the area of bate cells were greater in groups QLT,BWDHW and CLT than in the grouPmodel.CONCLUSION: Three formulas retard the development of hyperglycemia of rats induced with multiplelow doses STZ by increasing the cell mediated immunofunction.The formulas Ba-wei-di-huang-wan and Chailing-tang can reduce hyperglycemia.
