摘要/Abstract

摘要： 目的: 考察口服小剂量可溶性鸡Ⅱ 型胶原(SCC Ⅱ) 对大鼠佐剂性关节炎(AA) 的免疫治疗作用。方法: 检测脾细胞、肠系膜淋巴结细胞(MLNCs)和肠派伊尔结细胞(PPCs) 的ConA 增殖反应、腹腔巨噬细胞(PMΥ) 和滑膜细胞(SMCs) 产生IL-1 和TNFα水平, 结合观察踝关节继发炎症、胸腺指数等指标变化。结果: SCC Ⅱ(0.03 、0.30 和3.00 mg·kg^-1·d^-1,ig) 治疗(d 12 ~ 18), 能提升患鼠脾细胞低下的ConA增殖反应, 进一步抑制MLNCs 和PPCs 低下的ConA增殖反应, 显著抑制PMΥ和SMCs 产生IL-1 和TNFα, 明显减轻大鼠AA 的炎症反应。结论: 口服小剂量可溶性SCC Ⅱ对大鼠AA 有免疫治疗作用。

Abstract: AIM: To investigate the immune therapeutic effect of a small amount of oral soluble chicken collagen type Ⅱ (SCC Ⅱ) on adjuvant arthritis (AA) in rats.METHODS: The ConA-induced the proliferative responses of splenocytes, mesenteric lymph node cells(MLNCs) and peyer' s patche cells(PPCs), IL-1 and TNFαproductions in the supernatants of peritoneal macrophages(PMΥ) and synovial membrane cells (SMCs) combined with the changes of the indices such as ankle joint secondary inflammatory responses, thymus indices and so on in AA rats were assayed or observed.RESULTS: SCC Ⅱ(0.03, 0.30 and 3.00 mg·kg^-1·d^-1, d 12-18, ig) could improve the overreduced ConA-induced splenocyte proliferative responses, but further reduced those of MLNCs and PPCs, inhibited the IL-1 and TNFαproductions in PMΥand SMCs, and ameliorated the secondary inflammatory responses in AA rats.CONCLUSION: A small amount of oral SCC Ⅱ(ig) have the immunotherapeutic effect on AA in rats.
