水飞蓟宾对抗大鼠酒精性脂肪肝作用及机制

Effects ant mechanism of silibinin on antagonizing alcoholic fatty liver in rats

摘要: 目的: 观察水飞蓟宾(Silbinin)对大鼠酒精性脂肪肝作用及其机制。方法: 饮用10 %乙醇,加高脂高热量饲料喂饲6 周,造成大鼠酒精性脂肪肝模型;灌胃给药水飞蓟宾100mg·kg-1,继续酒精和高脂高热量饲料处理4 周后,处死大鼠,测定血清天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、碱性磷酸酶(AKP)活性和甘油三酯(TG)、胆固醇(TC)、低密度脂蛋白-胆固醇(LDL-C)、高密度脂蛋白-胆固醇(HDL-C)、大鼠肿瘤坏死因子-α(TNF-α)、大鼠转化生长因子-β1(TGF-β1)含量,测定肝细胞TG、肝匀浆丙二醛(MDA)、超氧化歧化酶(SOD)、还原性谷胱甘肽(GSH)水平以及肝组织学检查。结果: 水飞蓟宾能抑制病鼠血清AST、ALT、AKP活性(P<0.05或0.01)和降低TG、LDL-C、TNF-α、TGF-β1 含量(P<0.05或0.01),提高HDL-C 含量;降低肝匀浆TG 和MDA 含量(P<0.05或0.01),增强SOD 活性(P<0.01)以及改善肝细胞水变性和脂肪变性(P<0.01)。结论: 水飞蓟素阻止大鼠酒精性脂肪肝形成,其作用机制包括:抗氧化、清除自由基代谢产物,抑制脂质过氧化反应;调血脂、减少脂肪在肝脏的沉积;抗免疫性炎症和抗增殖。

Abstract: AIM: To investigate the effects and mechanism of silibinin(Sil)on rats with alcoholic fatty liver.METHODS: The animal models with alcoholic fatty liver were prepared in rats by feeding on high caloric food and drinking 10 % alcohol qd for 10 weeks ;after 6-week treatment, the effects of Sil (intragastrointestinally treated 100 mg·kg-1, qd for 4 weeks)were observed by serum activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (AKP), and serum contents of total cholesterols (TC), triacylglycerol (TG), low-density lipoprotein cholesterol (LDLC), high-density lipoprotein cholesterol (HDL-C), tumor necrosis factor (TNF)-αand transforming growth factor (TGF)-β1, and the contents of TG, malondialdehyde (MDA), glutathione (GSH), and activity of superoxide dismutase (SOD)in the hepatic tissue.Hepatic histopathology was observed by microscope.RESULTS: Serum levels of ALT, AST and AKP decreased in administration of Sil (P<0.05 or 0.01), the levels of TG, LDL-C, TNF-αand TGF-β1 reduced (P<0.05 or 0.01), the concentrations of TG and MDA in the hepatic tissue lowered (P<0.01), and the activity of SOD potentiated (P<0.01).Hydropic and or fatty degeneration of hepatocytes were improved (P<0.01).CONCLUSION: Silibinin can preclude alcoholic fatty liver in rats from drinking alcohol and feeding high caloric food, and pharmacological mechanisms were involved in anti-oxidation and removing free radical, inhibiting lipid peroxidation, adapting blood lipid component, reducing fatty sediment in liver, anti-inflammation and anti-hyperplasia.
