一种拟青霉代谢产物提取物对慢性应激抑郁大鼠血CORT、ACTH含量及下丘脑CRHmRNA表达的影响

Effects of BCPT on blood level of CORT and ACTH and expression of CRH MRNA of hypothalamuc in chronic stress rats

摘要: 目的: 研究一种拟青霉代谢产物提取物(bio-active compounds from paecilomyces tenuipes, BCPT)对慢性应激抑郁大鼠血皮质酮(CORT)、促肾上腺皮质激素(ACTH)含量及下丘脑促肾上腺皮质激素释放激素(CRH)mRNA表达的影响。方法: 在慢性温和性不可预见性应激(CUMS)模型上,采用糖水消耗量法观测动物的奖赏反应;以放射免疫分析方法(RIA)测定血清CORT及血浆ACTH含量;利用半定量逆转录-聚合酶链反应(RT-PCR)方法,测定下丘脑CRHmRNA的表达。结果: BCPT 40、80 mg·kg-1可不同程度增加模型大鼠糖水消耗量,改善模型大鼠的奖赏反应,并能不同程度降低模型大鼠血清CORT、血浆ACTH含量;下调慢性应激大鼠下丘脑CRHmRNA的过度表达。结论: BCPT的抗抑郁作用可能与其改善慢性应激大鼠下丘脑-垂体-肾上腺轴(HPAA)功能亢进有关。

Abstract: AIM: To study the effect of bioactive compounds from paecilomyces tenuipes (BCPT) on the blood level of CORT, ACTH and expression of CRH MR-NA of hypothalamus in chronic unpredictable mild stress model (CUMS) of depression in rats.METHODS: The epression animal model was induced by CUMS. The reward reaction was tested by the sweet water consumptionThe effect of BCPT on the blood level of CORT and ACTHwas tested by radioimmunoassay RT-PCR was used totest the expression of CRH MRNA of hypothalamus.RESULTS: BCPT (40,80 mg·kg-1) improved the reward re-action by increasing sweet water consumption, decreasing the expression of CRH MRNA of hypothalamus and the blood level of CORT and ACTH in chronic stressed ratsobviously.CONCLUSION: BCPT exhibited antidepres-sant-like effeet may in part be associated with regulating the hyperactingunction of hypothalamic-pitultaryadrenal axis (HPAA).
