苯并二氢吡喃衍生物 xy9902 对乳腺癌细胞增殖的影响

Effects of xy9902 a centchroman derivative on proliferation of MCF-7 cells

摘要: 目的: 研究苯并二氢吡喃衍生物 xy9902对乳 腺癌细胞增殖的影响。 方法: 采用MTT比色法测定化合物对MCF-7细胞的增殖作用;用流式细胞术测定化合物对细胞周期的影响;用放射性配体结合法 考察化合物与雌激素受体(estrogenic receptor, ER) 的 亲和力。 结果: 低浓度的 xy9902 促进 MCF-7 细胞增 殖并使细胞周期中 S 期细胞比例增多, 高浓度对 MCF-7 细胞增殖有抑制作用并使 S 期细胞比例减少;他莫昔芬可阻断 xy9902 对 MCF-7 细胞的增殖作 用;xy9902 对 ERα和 ERβ 的 IC50 分别为8.45 ×10-6 和 1.66×10-6 mol·L-1结论: 化合物 xy9902 低浓度促进 MCF-7 细胞增殖, 高浓度抑制 MCF-7 细胞增殖, 其增殖作用机制与 ERα激动有关。

Abstract: AIM: To investigate the effects of xy9902 on the proliferation in MCF-7 cells and its mechanism. METHODS: Effects of xy9902 on the proliferation of MCF-7 cellswere determined by MTT measurement.Cell cycle was examined by flow cytometry.Competitive estro-gen-receptor binding assay was used to investigate the af-finity of xy9902 to ER.RESULTS: xy9902 induced cell proliferation of MCF-7 cells at a lower concentration with an increase of the number of cells in S phase.On the contrary, inhibited cell proliferation at higher concentra-tionswith a decrease of the number of cells in S phase. Tamoxifen could block the stimulation of proliferation ef-fect of MCF-7 induced by xy9902.The relative binding affinity of xy9906 for ERαand ERβ showed an IC50 of 8.45 ×10-6 and 1.66 ×10-6 mol·L-1 respectively. CONCLUSION: xy9902 directly stimulates cell prolifer-ation of MCF-7 cells through ligand-receptor binding at a low concentrations and inhibits cell proliferation at high concentrations.
