鬼臼毒素纳米脂质体对人白血病细胞的凋亡诱导作用

Induction of apoptosis in human leukemia cells by podophyllotoxin nanoliposome

摘要: 目的 研究鬼臼毒素纳米脂质体对人白血病细胞K562 的凋亡诱导效应。方法 薄膜超声分散法制备鬼臼毒素纳米脂质体。不同浓度的鬼臼毒素和鬼臼毒素纳米脂质体分别处理K562 细胞,MTT 法检测细胞的增殖活性, 细胞形态学改变和Annexin Ⅴ/PI 染色检测细胞凋亡。结果 鬼臼毒素纳米脂质体对K562 细胞增殖的抑制作用明显高于鬼臼毒素,24 、48 、72 h 的半数抑制浓度(IC50) 分别为0.71 、0.02 、0.003 μg·ml-1, 而鬼臼毒素为1.50 、0.12 、0.05 μg·ml-1;0.01 μg·ml-1 鬼臼毒素纳米脂质体处理24 、72 h, Annexin Ⅴ/PI 染色K562 细胞凋亡率分别为91.1%和92.7%, 但72 h 时以晚期凋亡为主;而0.01 μg·ml-1鬼臼毒素处理的细胞凋亡率分别为63.4%和66.8%。鬼臼毒素纳米脂质体和鬼臼毒素处理后,K562 细胞出现皱缩、胞膜起泡、凋亡小体等典型的细胞凋亡形态学改变。结论 鬼臼毒素纳米脂质体较鬼臼毒素对白血病K562 细胞具有更强的增殖抑制和凋亡诱导效应。

Abstract: AIM: To study the apoptosis-inducing effect of podophyllotoxin nanoliposome on human leukemia K562 cells.METHODS: The podophyllotoxin nanoliposome was prepared using a dried film-ultrasonic procedure.K562 cells were treated with different concentration of podophyllotoxin (POD) and podophyllotoxin nanoliposome(POD-nlip) in vitro.The cell proliferating activity was assessed with an MTT assay, and cell morphology and Annexin Ⅴ/PI staining were employed to determine apoptosis in K562 cells.RESULTS: The inhibitory effect of POD-nliPon K562 cells was much higher than that of POD, the 50% inhibitory concentration (IC50) at 24 h,48 h and 72 h was 0.71, 0.02 and 0.003 μg·ml-1 , respectively,and 1.50, 0.12 and 0.05 μg·ml-1 respectivelyfor POD.After treatment with 0.01 μg·ml-1 PODnlipand POD for 24 h and 72 h, the apoptotic rates ofK562 cells detected by Annexin Ⅴ/PI staining were 91.1% and 92.7% for POD-nlip, and 63.4% and 66.8% for POD, respectively.At 72 h the cells mainly showed late apoptotic phenomenon.The cells showed typical apoptotic morphological changes in both POD-nlipand POD-treated cells.CONCLUSION: Compared with podophyllotoxin, podophyllotoxin nanoliposome has more significant inhibitory and apoptosis-inducing effect on human leukemia cell line K562 cells.
