摘要/Abstract

摘要： 目的: 研究肝素与大鼠肝星状细胞(HSC) 作用后瘦素与转化生长因子β₁ (TGF β₁) 表达的变化和意义。方法: 实验分组:肝素Ⅰ (10 μg/mL)、Ⅱ(100 μg/mL)、Ⅲ(1 000 μg/mL) 组, 加生理盐水为对照组, 均培养48 h 。培养终止后吸取上清液-20 ℃冰冻保存, ELISA 法检测其上清液瘦素与TGF β₁ 水平,MTT 法观察细胞增殖情况。结果: 肝素Ⅱ、Ⅲ组HSC 培养上清液瘦素水平均显著低于对照组(P<0.01);肝素各组TGF β₁ 水平和平均吸光度均显著低于对照组(P<0.01) 。结论: 大鼠HSC 在肝素作用下瘦素与TGF β₁ 分泌均受到抑制, 且星状细胞的增殖减少。

Abstract: AIM: To explore the effects of heparin on the expression of leptin and transforming growth factor β₁ (TGF β₁) in rat hepatic stellate cells(HSCs). METHODS: HSCs were divided as follows:heparin group Ⅰ(10 μg/mL), Ⅱ(100 μg/mL), Ⅲ(1 000 μg/mL), and control group.The incubation liquid was extracted after cultured 48 h and frosted in-20 ℃.Leptin and TGF β₁ were detected by ELISA, the HSC proliferation was measured by MTT assay. RESULTS: The levels of leptin in heparin group Ⅱ, Ⅲ were significantly lower than that in control group(P<0.01).The level of TGF β₁ and the mean absorbance in heparin groups were significantly lower than that in control group (P<0.01). CONCLUSION: Heparin can decrease expression of leptin and TGF β₁ in HSCs and inhibit proliferation of HSCs.
