羟基脲对大鼠Sertoli-germ 细胞共培养的影响

Effect of hydroxyurea on rat Sertoli- germ cell Co-culture

摘要: 目的:观察羟基脲(Hydroxyurea, HU) 对雄性大鼠Sertoli-germ 细胞共培养的细胞毒性作用。方法:在Sertoli-germ 细胞共培养模型中分别加入不同浓度HU(0、 10-3、 10-2、 10-1 mol/L) 后, 孵育96 h; 以及加入10-2 mol/L HU 后分别孵育24、 48、 72、 96 h 。结果:在各给药组, 96 h 时HU 导致germ细胞从Sertoli 细胞层的脱落增加, 呈现量效关系。在10-3 mol/L 以上浓度组, 细胞活性逐渐下降, 细胞脱落增加。同样, 随着给药时间的增加, germ细胞脱落率和乳酸脱氢酶(LDH) 的漏出逐渐增加, 细胞活性逐渐下降。结论:HU 对雄性大鼠睾丸Sertoli-germ 细胞共培养模型的germ 细胞具有明显毒性作用。

Abstract: AIM:to evaluate the cytotoxic effects of Hydroxyurea (HU) in vitro employing rat testicular Sertoli-germ cell coculture. METHODS:Cytotoxic changes induced by HU (0, 10-3, 10-2, 10-1mol/L)in mixed cultures of Sertoli and germ cells were seen at 96 h after treatment, respectively. And 10-2 mol/L HU were added to the culture, at 24, 48, 72 and 96 h after treatment, respectively. RESULTS:HU led to an increase in germ cell detachment from the Sertoli cell monolayer in a dose dependent manner. Detached cells were observed and the viability of detached cells was decreased in 10-3 mol/L and above treated groups. Substantial leakage of the cytosolic enzyme lactate dehydrogenase (LDH) was observed in the medium after 96 h HU treatment at concentrations of 10-2mol/L and above treated groups. The same, HU exposure led to an increase in germ cell detachment, the leakage of LDH, a decrease in cell viability from the Sertoli cell monolayer in a time dependent manner compared with the control culture. CONCLUSION:It is shown that HU exhibited an obvious toxicity to the germ cells of male rats in Sertoli cell germ cell co-culture model.
