红霉素对哮喘大鼠核因子NF-κB 及Th2 类细胞因子的影响

Effects of erycin on NF-κB and Th2 in rats with asthma

摘要: 目的 探讨红霉素对哮喘大鼠NF-κB 、Th2类细胞因子的影响。 方法 分离、培养正常对照组、哮喘组、红霉素处理组大鼠的外周血单核细胞(PBMCs)。分别采用ELISA 法检测细胞上清液中IL-4 、IL-5 、IL-13 浓度。逆转录聚合酶链反应(RTPCR)、Western blotting 技术检测PBMCs NF-κB mRNA、NF-κB 蛋白的表达。 结果 哮喘组PBMCs mRNA、NF-κB 蛋白表达水平及细胞上清液中IL-4 、IL-5 、IL-13 的浓度较正常对照组显著升高(P<0.05), 红霉素处理组NF-κB mRNA 、NF-κB 蛋白表达水平及细胞上清液中IL-4 、IL-5 、IL-13 的浓度较哮喘组显著降低(P<0.05)。通过直线相关分析发现, PBMCs NF-κB 蛋白与细胞上清液中IL-4 、IL-5 、IL-13 的表达水平之间均呈显著正相关(r 分别为0.78 、0.86 、0.71, P 均<0.05)。 结论 NF-κB可能参与支气管哮喘的病理生理过程。红霉素可能通过作用于NF-κB 这一靶点发挥其免疫调节作用。

Abstract: AIM: To explore the effects of erycin on the expression of cytokines such as NF-κB and Th2 in asthmatic rats. METHODS: The peripheral blood monouclear cells(PBMCs) were separated and cultured from rats in bronchial asthma, normal control and erycin-treatment groups.The concentrations of IL-4, IL-5 and IL-13 in plasma, the expressions of NF-κB mRNA and NF-κB protein in PBMCs were detected by enzyme linked immunosorbent assay (ELISA), reverse transcription polymerase chain reaction (RT-PCR), Western blotting, respectively. RESULTS: Compared with normal group the expressions of NF-κB mRNA and NF-κB protein in PBMCs and the concentrations of IL-4, IL-5 and IL-13 in supernatant were higher in asthmatic group (P<0.05).Compared with the asthmatic group, the above indexes were decreased significantly in erycin-treatment group (P<0.05).There were positive correlations between the expression of NF-κB protein in PBMCs and the concentrations of IL-4, IL-5 and IL-13 in supernatant (r =0.78, 0.86 and 0.71, P<0.05). CONCLUSION: NF-κB may play a role in patho-physiological course of asthma.Erycin could effectively treat asthma by inhibiting the expression of NF-κB.
