RhBLyS对AA大鼠MLNLs功能的影响及TACI-Ig的作用

RhBLyS influenced MLNLs function of rats with AA and the effect of TACI-Ig

摘要: 摘要:目的: 研究跨膜激活剂及钙调亲环素配体相互作用分子的免疫球蛋白(transmembrane activator or calcium-modulating cyclophylin ligand-interactor- immunoglobulin, TACI-Ig)对rhBLyS体外诱导佐剂性关节炎(adjuvant arthritis, AA)大鼠肠系膜淋巴结细胞(mesenteric lymph node lymphocytes, MLNLs)功能的影响。方法:无菌分离AA大鼠MLNLs,给予rhBLyS体外刺激细胞增殖,设置TACI-Ig五个浓度给药组(0.01, 0.1, 1.0, 10, 100 μg/ml)和一个阴性对照组IgG-Fc(10μg/ml)。MTT法检测大鼠MLNLs增殖;流式细胞仪检测T细胞主要亚群所占比例及活性变化;酶联免疫法检测细胞因子IL-17水平变化;Western blot法检测TACI蛋白表达水平。结果: TACI-Ig降低rhBLyS诱导的AA大鼠MLNLs过高的增殖能力;降低总Th细胞(CD3+/CD4+)和调节性T细胞 (CD4+/CD25+)的细胞百分含量,升高未致敏T细胞(CD4+/CD62L+)的细胞百分含量;降低AA大鼠MLNLs上清液中升高的IL-17水平;降低TACI蛋白表达。结论:TACI-Ig对MLNLs增殖活化、细胞因子分泌及受体蛋白表达均有抑制作用。为阐明TACI-Ig免疫抑制提供分子药理学依据。

Abstract: Abstract:Objective:To investigate the TACI - Ig on MLNLs of AA rats which induced by rhBLyS.Methods: MLNLs from AA rats were activated by rhBLyS in vitro. Set five concentrations of TACI-Ig (0.01, 0.1, 1.0, 10, 100 μg/ml) and a negative control group IgG Fc (10 μg/ml). Proliferations of MLNLs were assayed by MTT. Proportion of T cell subpopulation and activity change in MLNLs were analysed by flow cytometry.The level of IL-17 were measured by ELISA. The expression of TACI were investigated by western blotting. Results: RhBLyS can lead to proliferative accentuation in MLNLs and TACI-Ig significantly inhibit the rhBLyS stimulated proliferative reactions of lymphocytes.And TACI-Ig significantly decreased the enhanced total (CD3+CD4+ T cell) and activated (CD4+CD25+ T cell), enhanced the decreased unsensitized (CD4+CD62L+ T cell) on CD4+ T cell in MLN lymphocytes of AA rats.And TACI-Ig decreased IL-17 production and TACI expression.Conclusion: TACI-Ig has inhibitory action on both proliferation and activation of MLNLs and providing molecular pharmacology evidence for illuminating immunosuppressive action.
