姜黄素对大鼠局灶性脑缺血再灌注血管内皮生长因子和水通道蛋白4的影响

Effects of Curcumin on VEGF and AQP - 4 of rats with focal cerebral ischemia-reperfusion injury

摘要: 目的 初探姜黄素(Curcumin)对大鼠局灶性脑缺血再灌注后血管内皮生长因子(vascular endothelial growth factor,VEGF)、水通道蛋白4 ( aquapo rin - 4, AQP - 4)表达的影响。方法 将72只大鼠随机分为: 假手术组;缺血再灌注组;姜黄素治疗组(100,300 mg?kg-1)。采用线栓法闭塞大鼠大脑中动脉(MCA),制备大鼠局灶性脑缺血再灌注(MCAO)模型。再灌注48h,分别对各组大鼠进行神经功能缺损评分及其脑含水量的测定;检测血脑屏障通透性;ELISA法检测VEGF、AQP - 4的表达;运用HE染色观察其脑组织病理形态学。结果 姜黄素治疗组可显著降低脑含水量;减轻神经功能损伤和脑组织形态学改变;维持血脑屏障完整性;下调VEGF和AQP - 4的表达。在上述变化中,高剂量组表现最为明显。结论 姜黄素对脑缺血再灌注损伤具有保护作用,姜黄素的脑保护作用可能与其下调VEGF和AQP - 4的表达,改变血脑屏障(blood-brian barrier,BBB)的通透性有关。

Abstract: Objective To investigate the effect about vascular endothelial growth factor(VEGF)、aquapo rin – 4(AQP - 4)on cerebral ischemia-reperfusion injury in rats. Method Adult male SD rats were randomly divided into four groups: sham group; ischemia-reperfusion group; Curcumin - treatment groups: a dose of 100mg / kg; Curcumin - treatment groups: a dose of 300mg / kg.using Plug-line method make focal cerebral ischemia and reperfusion of middle cerebral artery occlusion model of rat. After 48h,Make Neurological deficit scores estimate, Measure Brain water content, Detect blood-brain barrier permeability, Using ELISA to detect the expression of VEGF and AQP - 4, Make HE to observe morphological changes of brain tissue.Result Curcumin can reduce brain water content; Improve the nerve damage symptoms of MCAO rat; Relieve brain morphological changes and BBB permeability; Reduce the expression of VEGF、AQP - 4. In the above changes, the high-dose group showed the most obvious.Conclusion Curcumin can protect cerebral ischemia-reperfusion, This effect may be regulated by Reducing the expression of VEGF and AQP – 4.
