法舒地尔联合西地那非治疗肺动脉高压大鼠疗效观察

Effect of fasudil combined with sildenafil in the treatment of pulmonary hypertension in rats

摘要:

目的:探讨法舒地尔联合西地那非治疗肺动脉高压大鼠的疗效。方法: 将100只大鼠随机分成A、B、C、D和E组,其中A组为空白对照组,B组为肺动脉高压模型组,C组在建模基础上给予法舒地尔治疗,D组在建模基础上给予西地那非治疗,E组在建模基础上给予法舒地尔联合西地那非治疗,比较5组大鼠的血压指标、右心室功能、右心室肥大以及微血管重建情况。结果: C组、D组和E组大鼠右心室收缩压(RVSP)均显著低于B组大鼠,E组大鼠RVSP显著低于C组大鼠(P<0.05);E组大鼠右心室舒张末期直径(RVEDD)和右心室收缩末期直径(RVESD)显著低于C组大鼠,D组和E组大鼠右心室缩短分数显著高于B组大鼠(P<0.05);E组大鼠右心室肥大指数(RVH)和右心室比体质量(RV/BW)显著低于C组和D组大鼠(P<0.05);E组大鼠肺动脉内膜增厚率显著低于B组、C组和D组大鼠(P<0.05),E组大鼠肺动脉ρ激酶活性显著低于B组、C组和D组大鼠(P<0.05)。结论:法舒地尔联合西地那非治疗能够显著改善肺动脉高压大鼠血流动力学指标以及右心室功能,缓解右心室肥大,有利于微血管重建,二者在调控ρ激酶活性,治疗肺动脉过程中可能存在协调作用。

Abstract:

AIM: To investigate the curative effect of fasudil combined with sildenafil in the treatment of pulmonary hypertension in rats.  METHODS: 100 rats were randomly divided into A, B, C, D and E group.Group A was control group, group B was set for the model of pulmonary hypertension, group C was given fasudil treatment on the basis of modeling. Group D received sildenafil treatment on the basis of modeling, group E was given fasudil combined with sildenafil treatment on the basis of modeling. The blood pressure, right ventricular function, right ventricular hypertrophy and vascular reconstruction were compared between the 5 groups. RESULTS:The RVSP of C group, D group and E group were significantly lower than that of B group;the RVSP of E group was significantly lower than that of rats in the C group (P<0.05); RVEDD and RVESD of group E were significantly lower than those in the C group, and right ventricular fractional shortening of D group and E group were significantly higher than that in the B group (P<0.05); RV/LV+S and RV/BW of the rats in group E were significantly lower than that of C group and group D (P<0.05); pulmonary artery intimal thickening in rats of group E was significantly lower than that of B group, C group and D group (P<0.05).Rho kinase activity in pulmonary artery of E group was significantly lower than that of B group, C group and D group (P<0.05). CONCLUSION: Fasudil combined with sildenafil treatment can significantly improve the hemodynamics of rats with pulmonary hypertension and right ventricular function as well as relieve right ventricular hypertrophy.It is beneficial to microvascular reconstruction, and there is a cooperative relationship between the two drugs in the regulation of Rho kinase activity and the treatment of pulmonary artery.
