Effects of new ATP-sensitive potassium channel pener iptakalim on Parkinsonian symptoms induced by haloperidol in rats

摘要: 目的:研究ATP 敏感性钾通道(ATP-sensitive potassium channels, KATP) 开放剂埃他卡林(Iptakalim, Ipt) 对由氟哌啶醇(haloperidol) 诱导产生的大鼠帕金森样症状的影响。方法:应用氟哌啶醇诱导大鼠帕金森样症状, 以左旋多巴(levodopa, L-DOPA) 为阳性对照, 观察不同剂量Ipt 对大鼠运动潜伏时间、行走时间以及直立和梳洗次数的影响。结果:Ipt 能显著改善氟哌啶醇引起的大鼠自发性活动减少;能缓解氟哌啶醇引起的大鼠全身僵硬, 并且随剂量的增加,Ipt 的作用逐渐增强。结论:KATP 通道开放剂Ipt 能够改善氟哌啶醇引起的大鼠帕金森样症状, 是一个极富治疗潜力的先导化合物。

Abstract: AIM: To investigate the effects of the new KATP channel opener, Iptakalim (Ipt), on the Parkinsonian symptoms induced by haloperidol in rats. METHODS: Using L-DOPA as the positive drug, the influences of Ipt on latency time, walking time, rearing and grooming of the parkinsonian-like rats induced by haloperidol were observed.RESULTS: Ipt ameliorated hypolocomotion and assuaged catalepsy induced by haloperidol in rats. Moreover, the effects of Ipt (0.5 mg·kg-1) were better than those of L-DOPA (100 mg·kg-1).CONCLUSION: Opening of KATP channels may play a key role in improving Parkinsonian symptoms induced by haloperidol in rats.
