Effects of dexamethasone on the expression of NF-κB and survivin in acute pancreatitis

Abstract:

AIM: To explore the effect of dexamethasone on the expression of NF-κB and survivin in pancreas in acute pancreatitis (AP). METHODS: AP animal model were established using caerulein and were divided into the saline group (control group), the AP group and dexamethasone group. The ELISA method was used to detect TNF-α in serum in each group. Real-time Q-PCR and Western blot were used to detect NF-κB, survivin, TNF-α mRNA and protein expression in pancreas in each group. RESULTS: The results of ELISA test showed that dexamethasone could significantly reduce the content of TNF-α in serum compared with the AP group. The results of qPCR and Western blot showed that dexamethasone could significantly reduce the expression of survivin, NF-κB, TNF-α mRNA and protein in pancreas compared with the AP group.CONCLUSION: Through inhibiting the NF-κB activation, dexamethasone could inhibit the expression and anti-apoptotic effect of survivin, promote apoptosis, reduce necrosis of pancreatic acinar cells, and inhibit the synthesis and release of TNF-α.
