Research on three fluorospectrophotometry methods for determination of concentration of adriamycin in tumor tissue

Abstract: AIM: To establish a simple and rapid fluorospectrophotometry method for the determination of concentration of adriamycin (ADR)in tumor tissue by comparing with three fluorospectrophotometry methods. METHODS: The concentrations of ADR in water and in tumor tissue were measured by three fluorospectrophotometry methods: n-butanol, silver nitrate and acidityisopropanol assay.The fluorescence spectrum curve, recovery and sensitivity were compared.RESULTS: The method of silver nitrate for determination of concentration of ADR in tumor tissue had low background tissue fluorescence.The linear range was 0.2-0.8 mg·L-1, the limit of detection was 0 .15 mg·L-1, and the recovery was over 97% by silver nitrate method .The method of acidity- isopropanol and n-butanol for determination of ADR in tumor-tissue fluorescence and low recovery which was <40 % by acidity-isopropanol assay and <50 % by n-butanol assay, respectively.CONCLUSION: The method of silver nitrate shows accurate, sensitive and reliable characterization for determination of concentration of ADR in tumor tissue.
