Effect of atorvastatin on intracellular cholesterol content and efflux of macrophages separated from diabetic hamster

Abstract: AIM: To examine the effect of atorvastatin on the intercellular cholesterol content and efflux of diabetic golden hamster models, and to further explore the mechanism about inhibition of atorvastatin on progress of atherosclerotic score.METHODS: We established diabetic golden hamstermodels.Peritoneal macrophages were isolated, purified and cultured in vitro.Then macrophages were incubated with atorvastatin of different concentrations in vitro.Intracellular cholesterol content was determined with combination of enzymatic method and HPLC before and after atorvastatin treatments.RT-PCR was carried out to examine the expression of ABCA1mRNA in macrophages.RESULTS: Atorvastatin, by cooperation with apoA1, can promote intracellular cholesterol efflux significantly, which may be mediated by activation of cholesterol ester hydrolase and by promotion of ABCA1 expression in macrophages.CONCLUSION: Atorvastatin can increase ABCA1 mediated intracellular cholesterol efflux.
