Effects of Dexamethasoneon apoptosis of HepG2 cells induced by Cisplatin and expression of Bcl-2 and Caspase-3

Abstract: AIM: To investigate the mechanism of Dexamethasone (DEX) on apoptosis of HepG2 cells induced by Cisplatin (DDP) and the expression of Bcl-2 and Caspase-3. METHODS: HepG2 and DDP were co-cultured, a serials of concentrations DEX were added into it, the expression of mRNA and protein of Bcl-2 and Capase-3 were analyzed separately by RT-PCR and Western blot,and the percentage of cell apoptosis was analyzed by FACS. RESULTS: With DEX increases in concentration, the expression of Bcl-2 was increased and expression of Caspase-3 was decreased correspondently, the rate of apoptosis of HepG2 descends clearly.CONCLUSION: DEX restrains apoptosis of HepG2 cells induced by Cisplatin (DDP) partly through Bcl-2 anti-apoptosis pathways, and Caspase-3 plays an important role in regulating the cell apoptosis.
