Protective effects of proanthocyanidins on the endoplasmic reticulum stress in PC12 cells induced by H2O2

Abstract: AIM: To study the protective effects of the proanthocyanidins on the endoplasmic reticulum stress in PC12 cells induced by H2O2. METHODS: The models of alzheimer ' s disease were established by treating PC12 cells with H2O2 (100 μmol/L)for 18 h.Before modeling for 30 minutes proanthocyanidins 40 mg/L, SB203580 10 μmol/L,Tempol 800 μmol/L were administrated respectively. The level of BiP/GRP-78 mRNA was detected by RT-PCR. The formation rate of reactive oxygen species(ROS)positive cells was detected with flow cytometry,the mean fluorecence was calculated. RESULTS: The level of BiP/GRP-78 mRNA and the formation rate of ROS were decreased in PC12 cells treated with H2O2; the level of BiP/GRP-78 mRNA and the formation rate of ROS were decreased(P<0.01) with proanthocyanidins treatment(40 mg/L)in PC12 cells treated with H2O2. CONCLUSION: Proanthocyanidins can ameliorate endoplasmic reticulum stress in PC12 induced bywith H2O2.
