Effects of insulin on expression of fibronectin, laminin and transforming growth factorβ1 in cultured neonatal rat cardiac fibroblasts

摘要: 目的: 研究胰岛素对心肌成纤维细胞胞外基质纤维连接蛋白(fibronectin, FN)、层粘连蛋白(laminin,LN) 及细胞因子转化生长因子β1 (TGFβ1 ) 表达的影响, 探讨其在心肌肥厚中的作用。方法: 乳鼠心肌成纤维细胞的培养并鉴定;用Streptavidin-Peroxidase(SP) 染色方法, 比较胰岛素实验组和对照组乳鼠心肌成纤维细胞FN、LN 及TGFβ1 的表达。结果: 培养的细胞经形态学观察和波形蛋白(vimentin) SP染色, 证实为心肌成纤维细胞;10-8 mol·L-1 胰岛素实验组细胞膜上FN 和细胞浆内TGFβ1 的表达明显强于对照组(P <0.01);LN 在培养的心肌成纤维细胞上基本不表达。结论: 在体外, 胰岛素具有促进心肌成纤维细胞胞外基质FN 堆积的作用, 可能参与了心肌肥厚的发生和发展, 其机制可能与胰岛素促进心肌成纤维细胞TGFβ1 的合成与分泌有关。

Abstract: AIM: To study the effects of insulin on expression of fibronectin (FN), laminin(LN) and transforming growth factor β1 (TGFβ1) in cultured neonatal rat cardiac fibroblasts, and its role in induction of cardiac hypertrophy. METHODS: The neonatal rat cardiac fibroblasts were cultured and identified;the expressions of FN, LN and TGFβ1 in experimental group (insulin treatment group) of neonatal rat cardiac fibroblasts were compared with those in control group respectively by Streptavidin- Peroxidase (SP) method. RESULTS: The cultured cells showed the characteristic of morphology and immunocytochemistry of cardiac fibroblasts;the expression of FN on the cell membrane of cardiac fibroblasts in 10-8 mol·L -1 insulin treatment group increased significantly in comparison with control group (P <0.05);LN was hardly expressed on the cultured cardiac fibroblasts, and there was no difference between two groups;the expression of TGFβ1 in the cytoplasm of cardiac fibroblasts in 10-8 mol·L -1 insulin treatment group increased markedly compared to the control group. CONCLUSION: Insulin can promote FN expression on cardiac fibroblasts in vitro and may be involved in the pathogenesis of cardiac hypertrophy.The promotion of insulin on the synthesis and excretion of TGFβ1 in cardiac fibroblasts may account for this phenomena.
