芸香苷对实验性急性胰腺炎的保护作用与抗氧化的关系

Relationship between protective effect and antioxidative action of rutosid on experimental acute pancreatitis in rats

摘要: 目的:探讨芸香苷(Rutoside, Ru) 对大鼠实验性急性胰腺炎(AP) 的保护作用与抗氧化作用的关系。方法:脱氧胆酸钠逆行胆胰管注射诱发大鼠AP 模型。观察造模后不同时间血清淀粉酶活性及胰腺组织病理组织学改变, 检测血清一氧化氮(NO)水平以及造模后24 h 胰腺组织中超氧化物歧化酶(SOD) 活性和丙二醛(MDA) 含量。结果:Ru(30, 60,120 mg·kg-1, sc) 3 个剂量可不同程度地降低模型大鼠血清淀粉酶活性, 改善胰腺组织病理组织学损伤, 恢复升高的血清NO 水平, 提高SOD 活性和减少MDA 生成。结论:Ru 对实验性急性胰腺炎具有一定的保护作用, 其作用机制可能与改善机体内源性抗氧化系统、提高血清NO 水平和清除氧自由基对胰腺组织的损伤有关。

Abstract: AIM: To study the protective effect and the antioxidative action of rutoside (Ru) on experimental acute pancreatitis (AP) in rats.METHODS: AP model was induced by retrograde injection of sodium deoxycholate into bilipancreatic duct in rats.The amylase (AMS) activity and the nitric oxide (NO) level in serum were measured at 6 and 12 h after the induction of AP. The superoxide dismutase (SOD) activity and the malondialdehyde (MDA) content in pancreas tissue were measured at 24 h after the induction.Meanwhile, the pathologic change of pancrease at 24 h after the induction was observed.RESULTS: Ru (30, 60, and 120 mg·kg-1, sc) could decrease the serum levels of AMS and NO significantly, attenuate the pathologic leision of pancrease, enhance the SOD activity and reduce the MDA production in the pancreas tissue.CONCLUSION: Ru has certain protective effect on AP in rats by administrating subcutaneously.The mechanisms may be due to strengthening endogenous antioxidative system and the ability of scavenging oxygen-derived free radicals (OFR), improving the histophysiology of AP and attenuating the injury of pancreas.
