穿琥宁对致热大鼠下丘脑组织中PGE2 和cAMP 含量的影响

Effects of chuanhuning injection on contents of PGE2 and cAMP in hypothalamus in fever rats

摘要: 目的: 探讨穿琥宁注射液的解热机制。方法: 用酵母混悬液复制大鼠发热模型, 观察穿琥宁注射液对大鼠体温的影响及其量效关系, 并采用放免法测定下丘脑中PGE2 和cAMP 含量。结果: 发热模型组在皮下注射酵母混悬液6 h 时, ΔT 及TRI6 明显高于对照组(P <0.01), 穿琥宁125、250 mg·kg-1组的ΔT 及穿琥宁250 mg·kg-1组的TRI6 明显低于发热模型组(P <0.01), 且穿琥宁250 mg·kg-1组的ΔT及TRI6 低于125 mg·kg-1组(P <0.01) 。发热模型组下丘脑中PGE2 和cAMP 的含量明显高于对照组(P <0.01), 而穿琥宁125、250 mg·kg-1组明显低于发热模型组(P <0.01), 且穿琥宁250 mg·kg-1组低于125 mg·kg-1组(P <0.05) 。ΔT 与下丘脑中PGE2和cAMP 的含量之间呈明显正相关(P <0.01) 。结论: 穿琥宁注射液可通过抑制下丘脑中PGE2 和cAMP含量的升高而发挥解热作用, 并存在量效关系。

Abstract: AIM: To investigate the antipyretic mechanisms of the chuanhuning injection in rats. METHODS: The fever model was made by suspension of yeast which was injected hypodermically to the rats.The contents of PGE2 and cAMP in hypothalamus were measured by radioisotope method.RESULTS: The maximal increment in body temperature (ΔT) and 6 h thermal response index(TRI6) in the model group were significantly higher than those in the control group (P <0.01), ΔT in chuanhuning 125 mg·kg-1group and 250 mg·kg-1group and TRI6 in chuanhuning 250 mg·kg-1group were lower than those in the model group(P <0.01), and ΔT and TRI6 in chuanhuning 250 mg·kg-1group were lower than those in chuanhuning 125 mg·kg-1group (P <0.01). The contents of PGE2 and cAMP in hypothalamus in model group were more higher than those in the control group (P <0.01), the contents of PGE2 and cAMP in the chuanhuning groups were lower than those in the model group (P <0.01), and the contents of PGE2 and cAMP in chuanhuning 250mg·kg-1group were lower than those in chuanhuning 125 mg·kg-1group (P <0.05).ΔT is positively correlative with the contents of PGE2 and cAMP in hypothalamus (P <0.01).CONCLUSION: The antipyretic mechanisms of chuanhuning injection may be related to its decreasing the contents of PGE2 and cAMP in hypothalamus, and the effects are dosage-dependence.
