正交实验建立蛇毒抗瘤蛋白生物活性测定方法

Determination of biological activity of atranticance by orthogonal test

摘要: 目的: 建立蛇毒抗瘤蛋白生物活性测定方法。方法: 用正交实验确定实验条件, 求细胞 50%死亡时的样品浓度, 即 IC 50 值。结果: 此法使细胞死亡率与样品 的 Log 浓 度具 有良好 的线 性关 系 (r =0.9515, P <0.01), 重复性实验结果:板内 RSD 值为12.3%, 板间 RSD 值为 16.6%;不同实验次数 RSD值为 17.8%, 灵敏度为 36 μg°L-1, 并能反应出蛇毒抗瘤蛋白的抗肿瘤特异性。结论: 此方法特异, 简便且具有良好的重复性, 可以作为蛇毒抗瘤蛋白生物活性的测定方法。

Abstract: AIM: To develop a method for determi-nation of the biological activity of atranticance. METHODS: The conditions of the experiment were determined by orthogonal test, then IC 50 which was the concentration of the sample when the 50% death of cells was tested. RESULTS: The mortality rate had a good correlativity with Log consistency of the sample, (r =0.9515, P<0.01). And the coefficients of variation of reproducibility of intra-, inter and different test assays were 12.3%, 16.6% and 17. 8%, respectively. CONCLUSION: Thismethod is superfine, simple and highly reproducible. It can be used for determining the biological activity of a-tranticance.
