盐酸埃他卡林对自发性高血压大鼠肾组织KATP通道基因表达的影响

Effects of iptakalim hydrohloride on KATP genes expression in renal tissues of spontaneously hypertensive rats

摘要: 目的: 从分子生物学水平探讨盐酸埃他卡林(iptakalim hydrohloride, Ipt) 对自发 性高血压大 鼠(spontaneously hypertensive rats, SHR) 肾组织 KATP 亚型表达的影响。方法: SHR 于第 12 周龄进入实验, 实验设 Ipt 1、3 和 9 mg°kg-1°d-13 个剂量组, 盐酸苯那普利(benazepril) 3 mg°kg-1°d-1治疗组及 SHR 空白对照组, 另设同周龄同种属正常血压大鼠(wistarKyoto rat,WKY) 为正常对照组, 灌胃给药每天 1 次,连续 12周, 观察盐酸埃他卡林对血压和肾组织 K ATP亚型表达的影响。结果: SHR肾组织SUR2、Kir6.1、Kir1.1 mRNA 表达较WKY 大鼠明显升高, 盐酸埃他卡林 1、3、9 mg°kg-1°d-13 个剂量组治疗后均可明显降低血压同时下调肾脏高表达的 Kir6.1、Kir1.1mRNA 水平, 而对 SUR2 表达无明显影响。结论: 盐酸埃他卡林对 SHR 降压治疗对肾脏的保护作用可能与其影响Kir6.1、Kir1.1 基因表达有关。

Abstract: AIM: To investigate the effects of iptakalim hydrohloride on KATP mRNA expression in renal tissues of spontaneously hypertensive rats. METHODS: SHRs at the age of 12-week-old were treated with Ipt 1, 3, and 9 mg°kg-1°d-1, benazepril 3 mg°kg-1°d-1 once a day for 12 weeks, respectively. The same aged WKY ratswere used as normal control. The effects of Ipt on BP and renal K ATP mRNA expression were detected by RT-PCR.RESULTS: mRNA expression level of SUR2、 Kir6.1 and Kir1.1 increased in SHR.After administra-tion of 1, 3, and 9 mg°kg-1°d-1 Ipt, the levels of BP were decreased, and the mRNA expression of Kir6.1 and Kir1.1were decreased. But there was no change in mR-NA expression of SUR2.In addition, there was no signif-icantly difference of mRNA expression of Kir6.2 among the SHR groups and the WKY group. CONCLUSION: The renal protective effects of Ipt may be related to regu-lation of genes expression of Kir6.1 and Kir1.1.
