蝮蛇毒蛋白提取物刺激糖作用的初步研究

Study on the effect of the Agkistrodon halys's venom-stimulated glucose uptake

摘要: 目的: 探讨蛇毒蛋白提取物刺激Hep G2 细胞的葡萄糖吸收及对糖吸收相关酶Akt 活性的影响。方法: 培养的Hep G2 细胞分成4 组:正常对照组、胰岛素组、蛇毒蛋白提取物组、胰岛素+蛇毒蛋白提取物组, 每组6 孔。用液闪计数法检测Hep G2 细胞对葡萄糖的吸收;用Western blot 法检测Akt 的活性变化。结果: 与正常对照组相比, 蛇毒蛋白提取物组可显著增加细胞对葡萄糖的吸收(P<0.01),并加强胰岛素刺激的葡萄糖吸收(P<0.01); 蛇毒蛋白提取物可激活Akt 的活性。结论: 蛇毒蛋白提取物具有刺激糖吸收以及加强胰岛素刺激的糖吸收的作用, 可作为糖尿病治疗的潜在药物。

Abstract: AIM: To investigate the effect of Agkistrodon halys's venom on stimulation glucose uptake and the effect of the Agkistrodon halys's venom on Akt activity in Hep G2 cells. METHODS: The Hep G2 cells were inoculated in the culture capsule. The Hep G2 cells were randomized into four groups(n =6 wells each): Control group, insulin group, Agkistrodon halys's venom group and insulin with Agkistrodon halys's venom group. Scintillation was used to detect 2-DG uptake in Hep G2 cells. The phosphorylation of Akt was tested using western blot. RESULTS: Compared with control cells, Agkistrodon halys's venom significantly stimulated the glucose uptake (P<0.01) and increased the insulin-stimulated glucose uptake(P<0.01) in Hep G2 cells. The venom evidently enhanced the activity of Akt inHep G2 cells. CONCLUSION: The venom stimulates the glucose uptake and increases the insulin-stimulated glucose uptake in Hep G2 cells, is a potential anti-hyperglycemic agent.
