阿托伐他汀对ApoE 基因缺陷小鼠心肌肥厚一级预防的实验研究

Primary prevention effect of atorvastatin on myocardial hypertrophy in ApoE-deficient (ApoE-/-) mice

摘要: 目的: 观察阿托伐他汀早期干预对胆固醇膳食喂养的ApoE 基因缺陷[ApoE(-/-) 小鼠心脏形态学和心肌间质成分的影响, 探讨阿托伐他汀对ApoE(-/-) 小鼠心肌损害的一级预防作用。方法: 16 只8 周龄雄性ApoE(-/-) 小鼠随机被分为两组:模型组和阿托伐他汀组(10 mg·kg-1·d-1 ), 每组8 只, 饲以胆固醇膳食饲料喂养至24 周龄, 相同周龄的C57BL 6J 小鼠设为对照。在24 周结束时处死小鼠。常规检测血浆胆固醇水平, 留取新鲜心脏组织测定TC 及NO 、超氧化物歧化酶(SOD) 、丙二醛(MDA);另取心脏组织固定, 石蜡切片, HE 染色观察各组小鼠心脏形态学的变化,Masson 染色观察心肌胶原改变。结果: 与模型组相比, 阿托伐他汀干预组ApoE (-/-) 小鼠血浆、心脏组织胆固醇和MDA 水平明显降低(P<0.05),NO 和SOD 水平明显增加(P<0.05), 心肌细胞直径和心肌胶原含量明显减少(P<0.05) 。结论: 阿托伐他汀早期干预西方膳食喂养的ApoE(-/-)小鼠, 可以降低胆固醇水平, 增强抗氧化能力, 减少胶原含量, 可能减轻心脏重构。

Abstract: AIM: To study the changes in myocardial cells and myocardial interstitial substances in ApoE-deficienct [ApoE(-/-) ] mice feeding with cholesterol food, and approach the primary prevention effects with atorvastatin on ApoE-deficienct ApoE(-/-) mice. METHODS: Sixteen male ApoE-deficienct mice of eight weeks feeding with high cholesterol food were randomly divided into two groups:model group (n=8 ), atorvastatin early intervention group (10 mg·kg-1 ·d-1) (n=8), nomal C57BL 6J mice of the same ages were set as the control group, feeding with high cholesterol animal food for 24 weeks and then were executed.The serum cholesterol/Level was detected with common practice method, the contents TC, NO, SOD infresh heart tissue were determined.Other portions of heart tissue were fixted, paraffin section, the changes of mice morphology and myocardial collagen in all groups were observed by HE and Masson dyeing, respectively.RESULTS: Compared with model group, the levels of ApoE-deficienct (ApoE-/-) mice blood plasma, heart tissue cholesterol, malondialdehyde significantly decreased, and the levels of NO, SOD significantly increased (P<0.05), themyocardial cells diameter and the content of myocardial collagen in ApoEdeficienct (ApoE-/-) mice significantly decreased in atorvastatin early intervention group (P<0.05 ).CONCLUSION: Atorvastatin could decrease the level of cholesterol, enhance the antioxygen ability, decrease the content of myocardial collagen, lessen cardiac remodeling to the early intervention (ApoE-/-) mice with cholesterol feed.
