阿托伐他汀对鼠脑缺血再灌注后P-选择素、人髓过氧化物酶的影响

Effects of atorvastatin on P-selectin and MPO in cerebral ischemia reperfusion rats

摘要: 目的:观察阿托伐他汀对P-selectin 表达及人髓过氧化物酶( MPO) 的影响。方法:将SD 大鼠随机分为假手术组( Sham) 、脑缺血再灌注组(CIR) 、阿托伐他汀组( ATO) 。阿托伐他汀采用灌胃的方式给药。CIR 组按制作模型后断头时程不同又分为6 、12 、24 、48 、72 h 共5 个亚组。阿托伐他汀组按给药时间不同分为给药4 周、2 周、3 d和造模后即时组4 个亚组, 每亚组分别于造模后12 、24 h 检测。断头取脑, 制作标本, 连续切片作P-selectin 免疫组化染色, 脑组织匀浆测定MPO 。结果:CIR 组可见P-selectin 阳性表达血管数6 h开始上升, 12 h 达到高峰, 至第3 天仍有少量表达, 阿托伐他汀进行干预后, P-selectin 表达明显下调( P <0.01), 阿托伐他汀组内两两比较差异亦有统计学意义( P <0.01);各CIR 组MPO 含量明显高于Sham 组( P <0.01), P-selectin 表达与MPO含量呈线性正相关, 给阿托伐他汀、阿司匹林干预后脑组织匀浆MPO 含量下降, 阿托伐他汀组内两两比较差异亦有统计学意义( P <0.05) 。结论:脑缺血再灌注后P-selectin 表达和MPO 含量明显升高, 使用阿托伐他汀干预后P-selectin 、MPO 含量明显下降。

Abstract: AIM: To observe the effects on the expressions of P-selectin and MPO induced by cerebral ischemia reperfusion injury in the rats after using atorvastatin.METHODS: SD rats were randomly divideded into sham operation group( Sham), cerebral ischemia reperfusion group( CIR) and atorvastatin group( ATO). Aatorvastatin was given via intragastrical.ATO group were divided into four weeks, two weeks, three days later and the immediate subgroup.Sham and CIR groups were divided into 6, 12, 24, 48, 72 hours subgroups, and were examined 12, 24 hours later.The expressions of P-selectin by immunohistochemistry technique and the contents of MPO in brain tissue homogenate were detected.RESULTS: The expressions of Pselectin were increased obviously at 6 h after ischemia reperfusion, the peak was at 12 h, then gradually were decreased, small amounts were expressed at 3 d.The expressions of P-selectin were down-regulation, after using atorvastatin, and there were remarkable differences in ATO group ( P <0.01).Compared with Sham group, the contents of MPO were increased remarkably in CIR group ( P <0.01).There was a linear positive correlation between the expressions of P-selectin and the contents of MPO.The contents of MPO in brain homogenate were decreased treated with atorvastatin and acenterine.There was statistical significance within ATO group( P <0.05).CONCLUSION: The expressions of P-selectin and the contents of MPO are increased after cerebral ischemia reperfusion, and decreased after using atorvastatin ( r =0.883, P < 0.05).
