N-乙酰基-L-半胱氨酸促进胰岛素经TR146细胞层渗透转运的研究

Effects of N-acetyl-L-cysteine on promoting permeation and transport of insulin across the buccal TR146 cell culture model

摘要: 目的:研究促吸收剂N-乙酰基-L-半胱氨酸(NAC)对胰岛素经TR146 口腔上皮细胞层渗透转运的影响。方法:应用TR146 细胞系, 采用Millicell 插入式细胞培养皿, 建立口腔上皮细胞渗透转运模型, 研究1.25 %、2.5 %、5 %、10 %各浓度NAC 对胰岛素经口腔上皮渗透转运的影响, 计算相应的胰岛素表观渗透系数Papp 和NAC 促渗比ER, 并用MTT 方法研究不同浓度NAC 对细胞活力的影响。结果:5 %、10 % NAC 可显著促进胰岛素经TR146 口腔上皮细胞层的渗透转运, 其Papp 分别为4.92 ±0.62 、5.04 ±0.52(×10-6 cms, n =5),ER 值分别为5.22 ±0.88 、6.05 ± 0.18(n =5), 明显高于对照组(P <0.05);10 %NAC组细胞相对酶活性显著下降, 而其余各组与对照组相比均无统计学差异。结论:5 %NAC 能显著促进胰岛素经TR146 口腔上皮细胞层的渗透转运, 且对细胞活力影响较小, 可作为一种安全有效的促吸收剂。

Abstract: AIM: To study the effects of N-acetyl-L-cysteine (NAC)on permeation and transport of insulin across the buccal TR146 cell culture model. METHODS: A human buccal epithelium model was established using TR146 cell line grown on Millicell culture plate inserts.The effects of NAC (1.25 %, 2.5 %, 5 %, 10 %)on permeation and transport of insulin across TR146 multilayer were studied.The apparent permeability coefficient (Papp)of insulin and the enhancement ratio (ER)of NAC were calculated, and the effects of NAC (1.25 %, 2.5 %, 5 %, 10 %) on TR146 cell activity were studied with MTT assay. RESULTS: 5 %, 10 % NAC significantly enhanced the permeation and transport of insulin across TR146 multilayer.Their Papp were 4.92 ±0.62 and 5.04 ± 0.52 (×10 -6 cm s, n =5) respectively, and ER were 5.22 ±0.88 and 6.05 ±0.18 respectively (n = 5), all of which were statistically higher than that of control group (P <0.05).The cell relative enzyme activity was significantly decreased, and those in the other groups had no significant defference compared with control group.CONCLUSION: 5 % NAC can greatly enhance the permeation and transport of insulin across TR146 multilayer, with little toxicity effects on the activity of TR146 cells.Therefore, NAC would be one safe and effective permeation enhancer for insulin transport.
