皖南尖吻蝮蛇毒抗凝蛋白组分的分离纯化及其急性毒性实验研究

Separation and purification of the anti-clotting protein component from venom of Agkistrodon and its acute toxicity testing

摘要: 目的 从皖南尖吻蝮蛇毒中分离纯化一种新的抗凝蛋白组分-7221(ACPF-7221)。研究小鼠对ACPF-7221 的急性毒性反应及死亡率并确定其LD50及95 %可信区间。 方法 先后用DEAE Sepharose Fast Flow 阴离子交换柱层析、Sephadex G75 分子筛、SP Sepharose Fast Flow 阳离子交换柱层析、Sephadex G50 分子筛过滤对尖吻蝮蛇粗毒进行分离纯化, 利用部分凝血活酶活性时间(APTT)活性检测筛选出抗凝成分。ACPF-7221 从小鼠的尾静脉注射, 根据每组小鼠的体重确定其相对应的给药量。根据死亡结果计算出LD50 及其95 %可信区间。 结果 分离纯化的ACPF-7221 由相对分子质量分别为25000 、30000 、50000 的3 种蛋白所构成三聚体, 其蛋白含量约为65 %。小鼠静脉注射LD50为7.848 mg kg, LD50(95 %可信区间)分别为7.353 ~ 8.375 mg kg。 结论 利用离子交换柱层析法可从皖南尖吻蝮蛇毒中提取出一种新的抗凝蛋白组分, 此组分由3 种相对分子质量各异的蛋白所构成三聚体。通过对小鼠急性毒性实验确定LD50及其95 %可信区间。肺为急性毒性的靶器官, 肺出血窒息是其死亡的原因。

Abstract: AIM: To separate and purify a new anti-clotting protein component(ACPF-7221)from the venom of Agkistrodon.The acute toxicity experiment of ACPF-7221 and the death rate were studied and the LD50 and 95 % confidence interval were determined in mice. METHODS: The venom of Agkistrodon was separated and purified using ion-exchange chromatography of DEAE-Sepharose Fast Flow, molecular sieve filtration through Sephadex G75, SP-Sepharose Fast Flow and molecular sieve filtration through Sephadex G50, the anticoagulation components were recalled using activated partial thromboplastin time (APTT)activity assay.ACPF-7221 was given to mice by intravenous injection, and the dosage was determined according to the body weight of mice.The LD50 and 95 % confidence were calculated by the death outcome. RESULTS: ACPF-7221 was separated and purified, which was a trimer consisted of three kinds of protein and their relative molecular mass were 25000, 30000 and 50000, and the protein level was 65 %.The LD50 of ACPF-7221 was 7.848mg kg, and its 95 % confident interval was 7.353-8.375 mg kg. CONCLUSION: A new anti-clotting protein component ACPF-7221 was extracted from the venom of Agkistrodon by ion-exchange chromatography, which is a trimer consisted of three kinds of protein.The critical orga of acute toxicity is lung, empsyxis asphyxia deduces death.
