AEG-1基因在肿瘤研究中的进展

Progresses of AEG-1 on cancer research

摘要: 星形细胞上调基因1(astrocyte elevated gene-1,AEG-1)是近年来发现的新的癌基因,该基因位于与多种恶性肿瘤发生密切相关的人类8号染色体(8q22)上,进一步研究表明其是促进肿瘤发生发展的一个关键因子。现已证实AEG-1是Ras/PI3K/Akt/c-Myc信号转导通路的下游靶分子,可以通过调控PI3K/Akt、NF-κB、Wnt/β-catenin、MAPK等多条信号途径参与正常细胞转化,肿瘤细胞增殖、侵袭、转移,血管生成等肿瘤发生发展的多个环节,并能介导肿瘤细胞自噬活性和化疗耐药。该基因对多种恶性肿瘤的诊断和预后具有重要的临床意义,可能成为肿瘤治疗和抗癌药物研发的新靶点。本文对近年来AEG-1在肿瘤研究中的进展作一综述。

Abstract: Astrocyte-elevated gene-1 (AEG-1), a novel oncogene, has been implicated in several crucial aspects of tumor progression, including oncogenic transformation, evasion of apoptosis, proliferation, invasion, metastasis, angiogenesis, chemoresistance and protective autophagy. Human AEG-1 gene is located in chromosome 8q22, which is known to be a hot spot for genomic alterations in several cancer cells. AEG-1 is regulated by Ras/PI3K/Akt/c-Myc pathway and exerts its effects by activating various pathways such as PI3K/Akt, NF-κB, Wnt/β-catenin, MAPK and so on. Since its clinical significance and biological role in human cancers, AEG-1 provides a novel target for intervening in the cancer phenotype. This article reviewed the current progress in the research field of AEG-1 in human tumor.
