Effects of anisodamine on vascular endothelium through blocking endothelial target for acetylcholine

Abstract: AIM: To investigate the change of endothelial structure and function by blocking endothelial target for acetylcholine with anisodamine.METHODS: Rabbits were killed after having been fed with anisodamine for three months.The morphological changes of vascular walls of aorta were observed through optical macroscope,and the morphological changes of vascular endothelial cells under electronic microscopes.The functions of aorta,pulmonary,carotid,renal and femoral vascular were measured by isolated vascular ring experiment.RESULTS: The aorta endothelial morphology of rabbits changed in treated with anisodamine for three months.The endothelial cells vertically adhered to internal elasticlamina.Some endothelial cells were mesh-shaped changed and some wereliable to desquamate.The endothelium -dependent vascular relaxation induced by acetylcholine was attenuated significantly.CONCLUSION: The vascular endothelial structure and function is damaged by blocking endothelial target for acetylcholine.
