Effects of novelPotassium channel openers aliphatic amines on hemodynamics in rats

Abstract: AIM: To study the effects of novelPotassium channel openers,aliphatic amines on hemodynamics in rats,and try to find out its structure-activity relationship.METHODS: After iv administration of 15 compounds at the dose of 5 mg·kg-1in anaesthetized rats,the bloodPressure was recorded byPolygraph with eight channels and the cardiac functionParameters by SMUPPC software to evaluate the effects of those compounds on hemodynamics.RESULTS: Among 15 compounds,the three compounds (No 4,No 7 and No 10)significantly decreased bloodPressure,slowed down heart rate,and inhibited the myocardial systolic and diastolic functions,butlowered theleft ventricular systolicPressure (lVSP)slightly.When the aliphatic amine has alkyl substituent with one to four carbons and has no cyclic substituent in its molecular structure,it showed strong effects on hemodynamics in rats.CONCLUSION: No 4,No 7 and No 10 of the compounds show strong effects on hemodynamics in anaesthetized normotensive rats,suggesting that those novelPotassium channel openers have strong cardiovascular effects.
