Inhibition of Cordyceps sinensis on gentamicin-induced apoptosis and bF-GF gene expression inProximal tubule epithelial cellline

Abstract: AIM: To investigate the effects of Cordycepssinensis in gentamicin-induced cytotoxicity,ap-optosis and basic fibroblast growth factor (bFGF)gene expression inlLCPK (aProximal tubule epithelial cellline).METHODS: Model grouPoflLCPK cells was stimulated with gentamicin,and at same time added Cordyceps sinensis in the treatment group.The number of cell survival was measured by MTT method,and NAGlevel in culture supernatant was detected by color reac-tion.Flow cytometry was used to detect the Annexin V upon the cytomembrane.The bFGF mRNA expression was quantitated by RT-PCR assay.RESULTS: After treat-ment with gentamicin,the number of cell survival de-creased,and NAGlevel increased.Gentamicin induced the apoptosis and high expression of bFGF mRNA inlL-CPK cells.Following incubation with Cordyceps sinensis,the number of cell survival increased,and thelevel NAG and apoptosis and bFGF mRNA expression decreased.CONCLUSION: Cordycepssinensis can decrease the cell damage reduced by gentamicin,and inhibits the apoptosis and bFGF mRNA expression inlLCPK cells.
