Regulatory effects of intracellular calcium in cultured endothelial cells through activating endothelial target for acetylcholine

Abstract: AIM: To examine intracellular calciumlevels in bovine aortic endothelial cells(BAEC)and human coronary aortic endothelial cells(HCAEC)by activating the endothelial target for acetylcholine(ETA).METHODS: As fluorescenceProbe,Fluo-3 was used in assaying [Ca2+]i in endothelial cells by cofocus.Nonselective agonists of cholinery receptors acetylcholine and carbachol were tested.RESULTS: Acetylcholine and carbachol can elicit the [Ca2+]i elevation between the 3rd to 10thPassage of BAEC or 8thPassage of HCAEC at the concentration between 10-5and 10-2mol·L-1.CONCLUSION: ETA can be activated by acetylcholine and carbachol and mediated the rise of intracellular Ca2 +in BAEC and HCAEC until the 10thPassage.This characteristic of ETA is notlost in culturedPassages.
