Modification of Centella asiatica compound on renal cytokines expressionProfiles in mice models with focal glomerulosclerosis

Abstract: AIM: To study the cytokine gene expressionProfile in mice model with focal glomerulosclerosis(FGS)nephritis and the mechanism of Centella asiatica compound on cytokines network.METHODS: FGS model mice of BALB/c were induced by adromycin(ADR,11mg·kg-1),and then treated with Centella asiatica compound for four weeks.The values of total RNA werePrepared from the kidney tissue at the end of first week and fourth week.The cDNA retained by the reversal react andlabeled with CY3-dUTP/CY5-dUTPin fluorescence,hybridized with the cytokines genes microarray.The chips were scanned in SCANARRAY 3000 and the images were analyzed by Image 4.0 software.RESULTS: The expression ofPDGF-α,PDGF-β,PDGF-R and VEGF mRNA were down-regulated after treated by Centella asiatica compound ;the expressions of IL-9,IL-7R2,and MIPwere down-regulated after treated by the compound.At the same time,through the stimulation of the compound,the expressions of c-Myc and Jun in kidney were restrained.CONCLUSION: Centella asiatica compound has a function of anti-glomerulosclerosis,and it may be a result of various cytokines worked together.
